The Adult, Larva, and Pupa of a New Pseudopyrochroa (Coleoptera: Pyrochroidae: Pyrochroinae) from China, with Molecular Phylogenetic Inferences.

A new species of Pseudopyrochroa Pic, 1906, P. reni Pan & Young, n. sp., is described from the western region of Mt. Qinling, China. Larvae, pupae, and adults were associated using molecular phylogenetic analyses based on mtDNA COI barcode sequences. All three stages are described and illustrated. Additionally, preliminary phylogenetic relationships among five genera and 14 species of Pyrochroidae, including Pseudopyrochroa, are hypothesized based on COI sequence data. The fauna of Pyrochroidae from the Mt. Qinling biodiversity conservation area is discussed.


First Descriptions of Larva and Pupa of Bagous claudicans Boheman, 1845 (Curculionidae, Bagoinae) and Systematic Position of the Species Based on Molecular and Morphological Data.

In this paper, the mature larva and pupa of Bagous claudicans are described and illustrated for the first time. Measurements of younger larval instars are also given. The biology of the species is discussed in association with larval morphology and feeding habits. Overall larval and pupal morphological characters of the genus Bagous are presented. Confirmation of the larva identification as Bagous claudicans species was conducted by cytochrome oxidase I (COI) sequencing. DNA barcoding was useful for specimen identification of larval stages. The systematic position of the species within the Bagous collignensis-group, based on morphological and molecular results, is also discussed.


The Adult and Larva of a New Species of the Genus Dila (Coleoptera, Blaptinae, Blaptini) from Himalayas, with Molecular Phylogenetic Inferences of Related Genera of the Blaptini.

In this study, a new species of the genus Dila Fischer von Waldheim, 1844, D. ngaria Li and Ren sp. n., was described from the southwestern Himalayas. The adult and larva were associated using molecular phylogenetic analyses based on fragments of three mitochondrial and one nuclear gene fragment (COI, Cytb, 16S and 28S-D2). Additionally, a preliminary phylogenetic tree was reconstructed and discussed based on a molecular dataset with seven related genera and 24 species of the tribe Blaptini. Meanwhile, the monophyly of the subtribe Dilina and the taxonomic status of D. bomina Ren and Li, 2001 are discussed. This work provides new molecular data for phylogenetic studies on the tribe Blaptini in the future.


Synthesis of Current Knowledge of the Morphology of the Larval Stages of Paederinae (Coleoptera; Staphylinidae), with a First Insight into the Mature Larva of Pseudomedon Mulsant & Rey, 1878, in the Light of a New Systematic Division.

This is the first morphological description of the mature larva (L2) of the genus Pseudomedon, belonging to the tribe Lathrobiini and subtribe Medonina. Detailed illustrations of its structural features are provided. Based on earlier published and new data, 10 and 18 diagnostic larval morphological characters for Paederinae and Pseudomedon, respectively, are proposed. In the light of the new tribal system established for the subfamily Paederinae and based on the current knowledge (including Pseudomedon) concerning the larval morphology of these staphylinids, a comparative analysis of the traits at different taxonomic levels was carried out: intertribal-between Lathrobiini and Paederini sensu nov., intersubtribal (Lathrobiini)-between Medonina and Lathrobiina, and intrasubtribal for Medonina. As a consequence, 12 intertribal, 2 intersubtribal and 3 intrasubtribal distinguishing features were selected. These features, appearing on the head, antennae and mouthparts of the larvae, confirm the validity of the recent proposals to alter the systematics of these staphylinids at higher taxonomic levels. Our proposed practical identification key to Paederinae larvae at the generic level is a synthesis of the current knowledge of Paederinae larvae, including new data. The work also gives a thorough review of the literature and the state of knowledge of the morphology of Paederinae larvae.


Cloak Scavenges the Reactive Oxygen Species around the Larvae of Drino inconspicuoides (Diptera: Tachinidae).

Drino inconspicuoides (Diptera: Tachinidae) is an endoparasitoid that develops inside the lepidopteran host. When the larva of D. inconspicuoides penetrates into the host, Mythimna separata (Lepidoptera: Noctuidae), the larva creates a cap-like structure, called the funnel, by using host hemocytes, forming a respiratory attachment to permit efficient respiration. A newly described cloudy and cottony structure, called the "cloak", is formed outside the funnel within 24 h of parasitism. The cloak contains the host fat body and hemocytes. In this study, we aimed to clarify the function of the cloak, which has to date remained unknown. We hypothesized that the funnel generates reactive oxygen species (ROS) through melanization, and that the cloak detoxifies them. We confirmed that the black pigments of the funnel were caused by melanization, which inevitably generates ROS that are potentially harmful to the D. inconspicuoides larva inside the funnel. The cloak showed high activities of antioxidant enzymes, including superoxide dismutase, glutathione peroxidase, and catalase. These results suggest that the cloak scavenged the ROS from the melanized funnel through the diversion of antioxidant enzymes in the fat body, thereby protecting the D. inconspicuoides larva from oxidative damage.


A New Species of Vietnamella Tshernova 1972 (Ephemeroptera: Vietnamellidae) from Thailand.

The larva, male subimago, female imago, and eggs of V. nanensis sp. n. are described based on specimens from Mae Hong Son and Nan provinces, Thailand. The female subimago is described based on a photograph of a specimen reared to the imago stage. The species previously was distinguished only by DNA barcode data and designated as Vietnamella sp. C. Based on morphology, the larva of the new species can be distinguished with the following combination of characteristics: (i) pattern of serration on the ventral margin of the forefemur, (ii) posterolateral margins of abdominal terga with pairs of acute tubercles, especially terga VI and VII, (iii) a well-developed pair of median ridge projections on tergum X, (iv) the second segment of the maxillary palp being about 1.3× the length of the third segment, and (v) females containing eggs with prominent protuberances on the chorionic surface. A key to larvae of all known species in the genus is provided.


Tolerance of High Oral Doses of Nonradioactive and Radioactive Caesium Chloride in the Pale Grass Blue Butterfly Zizeeria maha.

The biological effects of the Fukushima nuclear accident have been examined in the pale grass blue butterfly, Zizeeria maha (Lepidoptera: Lycaenidae). In previous internal exposure experiments, larvae were given field-collected contaminated host plant leaves that contained up to 43.5 kBq/kg (leaf) of radioactive caesium. Larvae ingested up to 480 kBq/kg (larva), resulting in high mortality and abnormality rates. However, these results need to be compared with the toxicological data of caesium. Here, we examined the toxicity of both nonradioactive and radioactive caesium chloride on the pale grass blue butterfly. Larvae were fed a caesium-containing artificial diet, ingesting up to 149 MBq/kg (larva) of radioactive caesium (137Cs) or a much higher amount of nonradioactive caesium. We examined the pupation rate, eclosion rate, survival rate up to the adult stage, and the forewing size. In contrast to previous internal exposure experiments using field-collected contaminated leaves, we could not detect any effect. We conclude that the butterfly is tolerant to ionising radiation from 137Cs in the range tested but is vulnerable to radioactive contamination in the field. These results suggest that the biological effects in the field may be mediated through ecological systems and cannot be estimated solely based on radiation doses.


In Vitro Effects of Pesticides on European Foulbrood in Honeybee Larvae.

Neonicotinoid and fungicide exposure has been linked to immunosuppression and increased susceptibility to disease in honeybees (Apis mellifera). European foulbrood, caused by the bacterium Melissococcus plutonius, is a disease of honeybee larvae which causes economic hardship for commercial beekeepers, in particular those whose colonies pollinate blueberries. We report for the first time in Canada, an atypical variant of M. plutonius isolated from a blueberry-pollinating colony. With this isolate, we used an in vitro larval infection system to study the effects of pesticide exposure on the development of European foulbrood disease. Pesticide doses tested were excessive (thiamethoxam and pyrimethanil) or maximal field-relevant (propiconazole and boscalid). We found that chronic exposure to the combination of thiamethoxam and propiconazole significantly decreased the survival of larvae infected with M. plutonius, while larvae chronically exposed to thiamethoxam and/or boscalid or pyrimethanil did not experience significant increases in mortality from M. plutonius infection in vitro. Based on these results, individual, calculated field-realistic residues of thiamethoxam and/or boscalid or pyrimethanil are unlikely to increase mortality from European foulbrood disease in honeybee worker brood, while the effects of field-relevant exposure to thiamethoxam and propiconazole on larval mortality from European foulbrood warrant further study.


Newly Emerging Pest in China, Rhynchaenusmaculosus (Coleoptera: Curculionidae): Morphology and Molecular Identification with DNA Barcoding.

The oak flea weevil, Rhynchaenusmaculosus Yang et Zhang 1991, is a newly emerging pest that severely damages oak (genus Quercus) in China. The first R. maculosus outbreak occurred in 2020 and caused spectacular damage to all oak forests in Jilin province, northeast China. The lack of key morphological characters complicates the identification of this native pest, especially in larva and pupa stages. This is problematic because quick and accurate species identification is crucial for early monitoring and intervention during outbreaks. Here, we provided the first detailed morphological description of R. maculosus at four life stages. Additionally, we used DNA barcodes from larva and pupa specimens collected from three remote locations for molecular identification. The average pairwise divergence of all sequences in this study was 0.51%, well below the 2% to 3% (K-2-parameter) threshold set for one species. All sample sequences matched the R. maculosus morphospecies (KX657706.1 and KX657707.1), with 99.23% to 100% (sequence identity, E value: 0.00) matching success. The tree based on barcodes placed the specimens into the Rhynchaenus group, and the phylogenetic relationship between 62 sequences (30 samples and 32 from GeneBank) had high congruence with the morphospecies taxa. The traditional DNA barcodes were successfully transformed into quick response codes with larger coding capacity for information storage. The results showed that DNA barcoding is reliable for R. maculosus identification. The integration of molecular and morphology-based methods contributes to accurate species identification of this newly emerging oak pest.


Identification of Chelonus sp. from Zambia and Its Performance on Different Aged Eggs of Spodoptera frugiperda.

The fall armyworm (FAW), Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae), is a migratory pest endemic, to tropical and subtropical regions of America. Biological control can effectively and sustainably control pests over a long period of time while reducing the frequency of pesticide use and ensuring the safety of agricultural produce. In our study, the egg-larval Chelonus species (Chelonus bifoveolatus) from parasitized eggs of Spodoptera frugiperda in Zambia were described and identified based on morphological and genetic characteristics. To evaluate the efficiency of C. bifoveolatus, their parasitism suitability on 0- to 2-day FAW eggs under laboratory conditions was compared. The results showed that C. bifoveolatus could accept all FAW eggs at 0-, 1- and 2-day-old age and complete development successfully. Significant differences were found among 0-, 1-, and 2-day-old host eggs with respect to egg-larva developmental duration of C. bifoveolatus, and the egg-larva developmental duration on 2-day-old eggs was significantly lower than those on 0- and 1-day-old eggs. No significant differences were observed in the parasitism, pupation, emergence, and female rates for C. bifoveolatus on various age eggs of FAW. Generally, the parasitism rate, pupal rate, and emergence rate at various ages of FAW eggs were higher than 90%, 75%, and 82%, respectively, and the longevity of female parasitoids was longer than male parasitoids, and the sex ratio of females to males was nearly 1:1. Our results indicate that C. bifoveolatus performed well on various ages of FAW eggs and is a potential biological control agent against FAW in Africa.


Host Immunosuppression Induced by Steinernema feltiae, an Entomopathogenic Nematode, through Inhibition of Eicosanoid Biosynthesis.

Steinernema feltiae K1 (Filipjev) (Nematode: Steinernematidae), an entomopathogenic nematode, was isolated and identified based on its morphological and molecular diagnostic characteristics. Its infective juveniles (IJs) were highly pathogenic to three lepidopteran (LC50 = 23.7-25.0 IJs/larva) and one coleopteran (LC50 = 39.3 IJs/larva) insect species. Infected larvae of the diamondback moth, Plutella xylostella (L.) (Insecta: Lepidoptera), exhibited significant reduction in phospholipase A2 (PLA2) activity in their plasma. The decrease of PLA2 activity was followed by significant septicemia of the larvae infected with S. feltiae. Insecticidal activity induced by S. feltiae was explained by significant immunosuppression in cellular immune responses measured by hemocyte nodule formation and total hemocyte count (THC). Although S. feltiae infection suppressed nodule formation and THC in the larvae, an addition of arachidonic acid (AA, a catalytic product of PLA2) rescued these larvae from fatal immunosuppression. In contrast, an addition of dexamethasone (a specific PLA2 inhibitor) enhanced the nematode's pathogenicity in a dose-dependent manner. To discriminate the immunosuppressive activity of a symbiotic bacterium (Xenorhabdus bovienii (Proteobacteria: Enterobacterales)) from the nematode, kanamycin was applied to after nematode infection. It significantly inhibited the bacterial growth in the hemolymph. Compared to nematode treatment alone, the addition of antibiotics to nematode infection partially rescued the immunosuppression measured by phenol oxidase activity. Consequently, treatment with antibiotics significantly rescued the larvae from the insecticidal activity of S. feltiae. These results suggest that immunosuppression induced by infection of S. feltiae depends on its symbiotic bacteria by inhibiting eicosanoid biosynthesis, resulting in significant insect mortality. However, the addition of antibiotics or AA could not completely rescue the virulence of the nematode, suggesting that the nematode itself also plays a role in its insecticidal activity.


Adult Postabdomen, Immature Stages and Biology of Euryommatus mariae Roger, 1856 (Coleoptera: Curculionidae: Conoderinae), a Legendary Weevil in Europe.

The larva and pupa of the saproxylic Euryommatus mariae Roger, 1857, the weevil species extremely rare in Europe, are described from Poland. It was reared from galleries in dead branches of a fallen spruce Picea abies. The larval morphology is compared with available larval descriptions of other genera of the supertribe Conoderitae, namely, the Palaearctic Coryssomerus, and the Nearctic Cylindrocopturus and Eulechriopus. The specific characters of the male and female postabdomen are described and illustrated, expressing the peculiar structure of endophallus and spermathecal duct, not seen in any other weevil species. A hypothesis regarding the mechanics of mating in this species is proposed. Euryommatus mariae is recorded for the first time to occur in China.


Micro-CT to Document the Coffee Bean Weevil, Araecerus fasciculatus (Coleoptera: Anthribidae), Inside Field-Collected Coffee Berries (Coffea canephora).

The coffee bean weevil, Araecerus fasciculatus (De Geer) (Coleoptera: Anthribidae), is a cosmopolitan insect with >100 hosts, and has been reported as a pest of stored coffee. During a study involving the coffee berry borer, we observed coffee bean weevils emerging from field-collected coffee berries and used micro-computerized tomography (micro-CT) scans to observe the insect inside the berry. Two eggs had eclosed inside the berry, resulting in observations of a newly eclosed adult beetle and a 5th instar larva, each feeding on one of the two seeds. This is the first time since 1775, when the insect was first described, that the insect has been observed inside a coffee berry.


Transcriptomic Analysis Insight into the Immune Modulation during the Interaction of Ophiocordyceps sinensis and Hepialus xiaojinensis.

Ophiocordyceps sinensis (Berk.) is an entomopathogenic fungus that can infect the larva of the ghost moth, Hepialus xiaojinensis, causing mummification after more than one year. This prolonged infection provides a valuable model for studying the immunological interplay between an insect host and a pathogenic fungus. A comparative transcriptome analysis of pre-infection (L) and one-year post-infection (IL) larvae was performed to investigate the immune response in the host. Here, a total of 59,668 unigenes were obtained using Illumina Sequencing in IL and L. Among the 345 identified immune-related genes, 83 out of 86 immune-related differentially expressed genes (DEGs) had a much higher expression in IL than in L. Furthermore, the immune-related DEGs were classified as pathogen recognition receptors (PRRs), signal modulators or transductors, and immune effector molecules. Serpins and protease inhibitors were found to be upregulated in the late phase of infection, suppressing the host’s immune response. Based on the above analysis, the expression levels of most immune-related genes would return to the baseline with the immune response being repressed in the late phase of infection, leading to the fungal immunological tolerance after prolonged infection. Meanwhile, the transcriptomes of IL and the mummified larva (ML) were compared to explore O. sinensis invasion. A total of 1408 novel genes were identified, with 162 of them annotated with putative functions. The gene families likely implicated in O. sinensis pathogenicity have been identified, primarily including serine carboxypeptidase, peroxidase, metalloprotease peptidase, aminopeptidases, cytochrome P450, and oxidoreductase. Furthermore, quantitative real-time PCR (qPCR) was used to assess the expression levels of some critical genes that were involved in immune response and fungal pathogenicity. The results showed that their expression levels were consistent with the transcriptomes. Taken together, our findings offered a comprehensive and precise transcriptome study to understand the immune defense in H. xiaojinensis and O. sinensis invasion, which would accelerate the large-scale artificial cultivation of this medicinal fungus.


BmAbl1 Regulates Silk Protein Synthesis via Glutathione Metabolism in Bombyx mori.

Bombyx mori, domesticated from wild silkworms, is an economic insect that feeds on mulberry leaves and produces silk. In the current study, we demonstrated the contribution of BmAbl1 in silk protein synthesis. The inhibition and knockout of BmAbl1 can reduce the larva weight and CSW. The effect on CSW of BmAbl1 is not on the transcriptional level, but on the translational level. RNA-sequencing data suggested that amino acid synthesis and the metabolism process had a great difference between the BmAbl1- and Control strain, particularly glutathione metabolism. An abnormality in glutathione metabolism led to the reduction of free glycine and serine content, which are the main components of fibroin protein. Finally, fibroin protein synthesis has been reduced, including fibroin-heavy chain, fibroin-light chain, and p25 protein. This finding brought to light the role of BmAbl1 in the silk protein synthesis process.


Developmental Biology and Identification of a Garden Pest, Otiorhynchus (Podoropelmus) smreczynskii Cmoluch, 1968 (Coleoptera, Curculionidae, Entiminae), with Comments on Its Origin and Distribution.

The mature larva and pupa of Otiorhynchus smreczynskii are described and illustrated with complete chaetotaxy for the first time. Five larval instars and the factors for larval growth are determined, and the larval development of this species is comprehensively described. In order to confirm species affiliation, selected larvae were subjected to genetic analysis (mtCOI). Host plants and unique feeding signs of some Entiminae species are presented, and all available data on development are documented and interpreted. Additionally, the morphometry of 78 specimens (48 of O. smreczynskii and 30 of O. rotundus) was examined in order to verify the usefulness of morphological features in distinguishing both species. The female genitalia of both species are illustrated, described and compared with each other for the first time. Finally, the updated distribution of O. smreczynskii is given, and a possible origin of O. smreczynskii and O. rotundus is proposed.


Subfamily Anischiinae (Coleoptera: Eucnemidae) in Early Cretaceous of Northeast China.

Rheanischia new genus, type species Rheanischia brevicornis new species (Eucnemidae, Anischiinae) is described from the Lower Cretaceous of Liaoning, China. The presence of this species in early Cretaceous deposits provides new insight into the evolution of basal lignicolous Eucnemidae clades. Both Anischiinae and Palaeoxeninae species diversified in a world dominated by gymnosperms, before the main radiation of angiosperms. More than 95% of modern eucnemid larvae have a Palaeoxenus-type highly modified head structure, but contrary to the Palaeoxenus larva, they develop in angiosperm wood. Anischiinae utilize angiosperms as well, but their head capsule shows no such modifications. These facts prove that highly specialized morphological features do not offer definite proof of similar way of life in the distant past, nor should non-modified structures be taken as proof for another kind of substrate choice. Eucnemidae have invaded angiosperms with two quite different morphological adaptations. This fact may have implications for the evolution of all clicking elateroids.


Impact of Sacbrood Virus on Larval Microbiome of Apis mellifera and Apis cerana.

In this study, we examined the impact of Sacbrood virus (SBV), the cause of larval honeybee (Apis mellifera) death, producing a liquefied a larva sac, on the gut bacterial communities on two larval honeybee species, Apis mellifera and Apis cerana. SBV was added into a worker jelly food mixture and bee larvae were grafted into each of the treatment groups for 24 h before DNA/RNA extraction. Confirmation of SBV infection was achieved using quantitative reverse transcription polymerase chain reaction (RT-qPCR) and visual symptomology. The 16S rDNA was sequenced by Illumina sequencing. The results showed the larvae were infected with SBV. The gut communities of infected A. cerana larvae exhibited a dramatic change compared with A. mellifera. In A. mellifera larvae, the Illumina sequencing revealed the proportion of Gilliamella, Snodgrassella and Fructobacillus was not significantly different, whereas in A. cerana, Gilliamella was significantly decreased (from 35.54% to 2.96%), however, with significant increase in Snodgrassella and Fructobacillus. The possibility of cross-infection should be further investigated.


Gamma Radiation Sterilization Dose of Adult Males in Asian Tiger Mosquito Pupae.

The pathogen-carrying tiger mosquito, Aedes albopictus, has spread from the Western Pacific and Southeast Asia to Europe, Africa, the Middle East, North and South America, and the Caribbean. This species of mosquito transmits arboviral infections, such as yellow fever, chikungunya, dengue, zika, and several encephalitides. The objective of this research was to provide a radiation dose inducing sterilization in adult male Ae. albopictus in the pupal stage. A cobalt-60 source of gamma radiation at a dose rate of 381 Gy/h was used. The pupae were irradiated with doses of 0 (control), 20, 30, 40, 50, and 60 Gy. Each treatment had a total of five replications using 60 pupae. After irradiation, the different phases of Ae. albopictus development (egg, larva, pupa, and adult) in the F1 generation were observed daily. Parameters such as viability, fertility, longevity, and mortality were recorded. The results from these studies showed that a dose of 60 Gy was necessary to sterilize 100% of the male Ae. albopictus pupae.


DNA Barcoding Identifies Unknown Females and Larvae of Fannia R.-D. (Diptera: Fanniidae) from Carrion Succession Experiment and Case Report.

Application of available keys to European Fanniidae did not facilitate unequivocal species identification for third instar larvae and females of Fannia Robineau-Desvoidy, 1830 collected during a study of arthropod succession on pig carrion. To link these samples to known species, we took the advantage of molecular identification methods and compared newly obtained cytochrome oxidase subunit I (COI) barcode sequences against sequences deposited in reference databases. As an outcome of the results obtained, we describe for the first time a third instar larva of Fannia nigra Malloch, 1910 and Fannia pallitibia (Rondani, 1866) and a female of Fannia collini d'Assis-Fonseca, 1966. We provide combinations of characters allowing for discrimination of described insects from other Fanniidae. We provide an update for the key by Rozkošný et al. 1997, which allows differentiation between females of F. collini and other species of Fanniidae.
