Oviposition, Feeding Preferences and Distribution of Delia Species (Diptera: Anthomyiidae) in Eastern Canadian Onions.

Delia antiqua, Delia platura and Delia florilega are three root maggot species commonly considered pests in Eastern Canadian onions. The onion maggot, D. antiqua, is considered the primary root maggot pest in onion but it remains unclear whether the other two species are also causing damage. In order to develop updated management strategies for root maggot, we tested adult oviposition and feeding preference by Delia larvae on four growth stages of onion using bioassays and we determined the Delia species composition across the four major onion growing regions in eastern Canada. Delia species oviposit readily on onion at the 5-7 true leaf growth stage but damage on onions is not statistically different between Delia species in our zero-inflated models. The four eastern Canadian onion growing regions have different proportions of Delia species. Southern Ontario and Quebec were the only two regions where Delia antiqua was collected. The highest average numbers of Delia spp. were caught in Quebec and Nova Scotia. Our study shows that timing is important in implementation of management strategies for root maggot in Eastern Canadian onions.


Leaf UV-B Irradiation and Mycorrhizal Symbionts Affect Lettuce VOC Emissions and Defence Mechanisms, but Not Aphid Feeding Preferences.

Arbuscular mycorrhizal fungi (AMF) and ultraviolet-B radiation (UV-B) play important roles in plant-insect interactions by altering plant physiology and histology. We hypothesized that UV-B-induced oxidative stress was mitigated by AMF symbiosis. In this study, we conducted a multifactorial experiment to explore lettuce plant response to AMF inoculation and UV-B exposure (0.4 W m-2; 16 h d-1; 2 weeks), either together or individually, as well as the interaction with the polyphagous insect pest Myzus persicae (Sulzer). Lettuce plants subjected to UV-B radiation showed an increase in callose and oxidative stress indicators, as well as a decrease in stomatal density. Mycorrhizal colonization cancelled out the effect of UV-B on stomatal density, while the symbiosis was not affected by UV-B treatment. The plant volatile emission was significantly altered by UV-B treatment. Specifically, the non-terpene 1-undecene abundance (+M/+UVB: 48.0 ± 7.78%; -M/+UVB: 56.6 ± 14.90%) was increased, whereas the content of the non-terpene aldehydes decanal (+M/+UVB: 8.50 ± 3.90%; -M/+UVB: 8.0 ± 4.87%) and undecanal (+M/+UVB: 2.1 ± 0.65%; -M/+UVB: 1.20 ± 1.18%) and the sesquiterpene hydrocarbons (+M/+UVB: 18.0 ± 9.62 %; -M/+UVB: 19.2 ± 5.90%) was decreased. Mycorrhization, on the other hand, had no significant effect on the plant volatilome, regardless of UV-B treatment. Aphid population was unaffected by any of the treatments, implying a neutral plant response. Overall, this study provides new insights about the interactions among plants, UV-B, and AMF, outlining their limited impact on a polyphagous insect pest.


DNA Metabarcoding as a Tool for Disentangling Food Webs in Agroecosystems.

Better knowledge of food webs and related ecological processes is fundamental to understanding the functional role of biodiversity in ecosystems. This is particularly true for pest regulation by natural enemies in agroecosystems. However, it is generally difficult to decipher the impact of predators, as they often leave no direct evidence of their activity. Metabarcoding via high-throughput sequencing (HTS) offers new opportunities for unraveling trophic linkages between generalist predators and their prey, and ultimately identifying key ecological drivers of natural pest regulation. Here, this approach proved effective in deciphering the diet composition of key predatory arthropods (nine species.; 27 prey taxa), insectivorous birds (one species, 13 prey taxa) and bats (one species; 103 prey taxa) sampled in a millet-based agroecosystem in Senegal. Such information makes it possible to identify the diet breadth and preferences of predators (e.g., mainly moths for bats), to design a qualitative trophic network, and to identify patterns of intraguild predation across arthropod predators, insectivorous vertebrates and parasitoids. Appropriateness and limitations of the proposed molecular-based approach for assessing the diet of crop pest predators and trophic linkages are discussed.


CCHamide-2 Signaling Regulates Food Intake and Metabolism in Gryllus bimaculatus.

Neuropeptides play vital roles in energy homeostasis in insects and other animals. Although the importance of the regulatory network of neuropeptides in feeding and metabolism has been illuminated, a complete understanding of the mechanisms has not been addressed as many factors are involved in the regulation. CCHamide-2 is a newly identified brain-gut neuropeptide that regulates feeding behavior in several insect species including Drosophila melanogaster. However, little is known about the mechanisms controlling the feeding-related behavior and metabolic functions modulated by CCHamide-2 in other insects. In this study, we addressed the functions of CCHamide-2 in the two-spotted cricket, Gryllus bimaculatus, which was used as the experimental material to research the mechanisms of feeding and metabolism in this omnivorous insect species. Knockdown crickets by RNA interference against GbCCHamide-2R increased the amount of food intake, while injection of chemically synthetic GbCCHamide-2 peptide reduced the amount of food intake. Further, knockdown and peptide injection experiments revealed that GbCCHamide-2 signaling increased the concentrations of circulating lipids and carbohydrates, and the carbohydrate-rich diet increased the transcript levels of GbCCHa-2R. Moreover, GbCCHa-2 injection decreased the transcript level of Gbilp. By contrast, GbCCHamide-2 signaling did not affect nymphal growth or the transcript level of GbAKH, as well as feeding preferences. Taken together, CCHamide-2 signaling in G. bimaculatus regulates food intake associated with alterations in lipid and carbohydrate levels in hemolymph.


Bumble Bee Foraged Pollen Analyses in Spring Time in Southern Estonia Shows Abundant Food Sources.

Agricultural landscapes usually provide higher quantities of single-source food, which are noticeably lacking in diversity and might thus have low nutrient value for bumble bee colony development. Here, in this study, we analysed the pollen foraging preferences over a large territory of a heterogeneous agricultural landscape: southern Estonia. We aimed to assess the botanical diversity of bumble bee food plants in the spring time there. We looked for preferences for some food plants or signs of food shortage that could be associated with any particular landscape features. For this purpose, we took Bombus terrestris commercial hives to the landscape, performed microscopy analyses and improved the results with the innovative DNA metabarcoding technique to determine the botanical origin of bumble bee-collected pollen. We found high variability of forage plants with no strong relationship with any particular landscape features. Based on the low number of plant species in single flights, we deduce that the availability of main forage plants is sufficient indicating rich forage availabilities. Despite specific limitations, we saw strong correlations between microscopy and DNA metabarcoding data usable for quantification analyses. As a conclusion, we saw that the spring-time vegetation in southern Estonia can support bumble bee colony development regardless of the detailed landscape structure. The absence of clearly dominating food preference by the tested generalist bumble bee species B. terrestris makes us suggest that other bumble bee species, at least food generalists, should also find plenty of forage in their early development phase.


Effects of Variety and Grape Berry Condition of Vitis vinifera on Preference Behavior and Performance of Drosophila suzukii.

Drosophila suzukii is an invasive fruit pest and represents a potential economic threat to viticulture. After first observations of D. suzukii in Europe in 2008, research mainly focused on the evaluation of the host range and infestation risk for fruit and berry crops. However, the risk assessment of D. suzukii in viticulture has only recently started. Understanding the factors influencing preferences of D. suzukii for host species and varieties as well as offspring performance is essential to improve management strategies. We investigated the field infestation of different grape varieties across Baden-Wuerttemberg, southwestern Germany, between 2015 and 2018. Moreover, we performed dual-choice assays in the laboratory to investigate whether adults show preferences for certain varieties and whether offspring performance differs between varieties. Furthermore, we studied the impact of grape damage on choice behavior. Field monitoring revealed that D. suzukii show preferences for red varieties, whereas almost no oviposition occurred in white varieties. The results of dual-choice assays confirmed that D. suzukii preference and performance are influenced by grape variety and that flies preferred damaged over intact "Pinot Noir", "Pinot Blanc", and "Müller-Thurgau" berries. Overall, these findings may have important implications for winegrowers regarding cultivated varieties, grape health, and insecticide reduction.


Exploring the Acceptance of Entomophagy: A Survey of Italian Consumers.

Insect-based food is not common in Europe, because most people do not consider insects to be edible, but rather a threat and a health risk. Fear and refusal to eat a new food product introduced into a culture is called food neophobia, which results in a hesitation to trying and experimenting with new foods. Although there is significant interest in this novel sector, there is a lack of research on the link between rejection, the level of food neophobia, and consumer behavior related to the introduction of insects into the diet. In this study, through 420 questionnaires, a specific experimental scale of insects was introduced which, together with a neophobia scale, analyzed the probability and the intention of respondents to consume insects. Another issue tested has been their intention to eat food containing insects. We observed that the analyses of the two scales produced different results, confirming the need for a specific scale to measure "insect phobia". This is important, since knowledge about consumer preferences for and barriers to using insects as human food sources is limited but necessary in order to set up commercialization strategies. The development of insect-based food offers physical health benefits and also improves the sustainability of the food industry.


Factors Influencing Consumption of Edible Insects for Chinese Consumers.

Edible insects are often considered a healthier and more sustainable meat substitute and protein source. Many studies have examined factors affecting the consumption behavior towards edible insects among Western consumers. However, little is known about factors influencing consumer behavior towards edible insects in Asian countries even though Asians have a long history of consuming insects. In this study, we surveyed 614 Chinese consumers from Beijing and Nanjing to examine the factors influencing their consumption and purchase behavior of edible insects. We find that insect phobia, feelings of disgust, knowledge level, and social demographic factors such as age, household size, household income and region (Northern or Southern China) are the main factors influencing purchase decisions. In addition, the results indicate that the perceived positive attributes associated with edible insects, the preferences of children in the household, as well as age and knowledge level have positive impacts on consumption frequency. On the other hand, concerns of food safety and the shape of the insects have negative impacts on consumption frequency. Finally, the results suggest that educating consumers to increase knowledge of edible insects increases their probability to purchase insect foods.


Colour Response in Western Flower Thrips Varies Intraspecifically.

Discrepancies in the published research as to the attraction of the economically important pest western flower thrips (WFT) to different colours confounds the optimisation of field traps for pest management purposes. We considered whether the different experimental conditions of independent studies could have contributed to this. Therefore, the behavioural response (i.e., landings) to different colour cues of two WFT laboratory populations from Germany (DE) and The Netherlands (NL), which had previously been independently shown to have different colour preferences, were tested in the same place, and under the same experimental conditions. Single-choice wind tunnel bioassays supported previous independent findings, with more of a NL population landing on the yellow LED lamp (588 nm) than the blue (470 nm) (p = 0.022), and a not-statistically significant trend observed in a DE population landing more on blue compared to yellow (p = 0.104). To account for potential original host rearing influences, both populations were subsequently established on bean for ~20 weeks, then yellow chrysanthemum for 4−8 and 12−14 weeks and tested in wind tunnel choice bioassays. Laboratory of origin, irrespective of the host plant rearing regime, remained a significant effect (p < 0.001), with 65% of the NL WFT landing on yellow compared to blue (35%), while 66% of the DE WFT landed on blue compared to yellow (34%). There was also a significant host plant effect (p < 0.001), with increased response to yellow independent of laboratory of origin after rearing on chrysanthemum for 12−14 weeks. Results suggest that differing responses of WFT populations to colour is, in this case, independent of the experimental situation. Long-term separate isolation from the wild cannot be excluded as a cause, and the implications of this for optimising the trap colour is discussed.


Influence of Microclimate Factors on Halyomorpha halys Dehydration.

Understanding the interaction between insects and microclimate can be essential in order to plan informed and efficient treatments against agricultural pests. Microclimatic factors such as humidity and temperature can influence the population dynamics of the invasive agricultural pest Halyomorpha halys, the brown marmorated stink bug. The aim of this work was to evaluate the level of transpiration of H. halys in dry, normal and humid microclimates according to the sex, physiological conditions and developmental stage of individuals. Water loss during diapause and the effect of population density on insects' transpiration were also assessed, as were the nutritional preferences of adults upon exiting diapause. Our data demonstrate that microclimatic conditions significantly influence the transpiration of this pest species. The effect of sex and feeding status on insects' water loss is marked, while population density does not influence water loss in diapausing individuals. The first nutritional need of the overwintering generations is represented by hydration, likely due to the water loss during diapause.


Hermetia illucens (L.) (Diptera: Stratiomyidae) Odorant Binding Proteins and Their Interactions with Selected Volatile Organic Compounds: An In Silico Approach.

The black soldier fly (BSF), Hermetia illucens (Diptera: Stratiomyidae), has considerable global interest due to its outstanding capacity in bioconverting organic waste to insect biomass, which can be used for livestock, poultry, and aquaculture feed. Mass production of this insect in colonies requires the development of methods concentrating oviposition in specific collection devices, while the mass production of larvae and disposing of waste may require substrates that are more palatable and more attractive to the insects. In insects, chemoreception plays an essential role throughout their life cycle, responding to an array of chemical, biological and environmental signals to locate and select food, mates, oviposition sites and avoid predators. To interpret these signals, insects use an arsenal of molecular components, including small proteins called odorant binding proteins (OBPs). Next generation sequencing was used to identify genes involved in chemoreception during the larval and adult stage of BSF, with particular attention to OBPs. The analysis of the de novo adult and larval transcriptome led to the identification of 27 and 31 OBPs for adults and larvae, respectively. Among these OBPs, 15 were common in larval and adult transcriptomes and the tertiary structures of 8 selected OBPs were modelled. In silico docking of ligands confirms the potential interaction with VOCs of interest. Starting from the information about the growth performance of H. illucens on different organic substrates from the agri-food sector, the present work demonstrates a possible correlation between a pool of selected VOCs, emitted by those substrates that are attractive for H. illucens females when searching for oviposition sites, as well as phagostimulants for larvae. The binding affinities between OBPs and selected ligands calculated by in silico modelling may indicate a correlation among OBPs, VOCs and behavioural preferences that will be the basis for further analysis.
