In the USA, lesbian, gay, bisexual, transgender and queer (LGBTQ+) people report higher rates of tobacco use than non-LGBTQ+ people due to diverse factors, from anti-LGBTQ stigma to targeted marketing by the tobacco industry. There is an opportunity to support behavioural changes by delivering evidence-based health communication campaigns through community-based organisations (CBOs), but an insufficient evidence base and organisational resource restrictions limit this potential. Our previous research with CBO staff and leaders serving LGBTQ+ communities identified gaps in evidence-based campaigns, such as insufficient centering of LGBTQ+ communities in campaign development and execution. This qualitative study explores opportunities to improve the products and services supporting the dissemination of evidence-based health communication campaigns for LGBTQ+ audiences.

Racial discrimination is a chronic stressor that may contribute to cardiovascular disease (CVD) disparities in non-Hispanic Black (NHB) adults. NHB adults experience greater perceived discrimination (PD) and CVD risk compared with non-Hispanic White adults. Although the association between PD and some subclinical CVD markers has been examined, the relationship between PD and arterial stiffness has not been established, limiting our understanding of the impact of PD on CVD.

Obesity (OB) in children remains a national epidemic. This trial targets children suffering from overweight (OW) and OB living in rural Appalachia. Rural Appalachia is fraught with unhealthy behaviours, high rates of OB, pre-diabetes and type 2 diabetes among children. Diabetes prevalence in Appalachia is nearly double that of other regions. The prevalence of Appalachian children suffering from OB and extreme obesity (EO) increases the severity of diabetes.

Ethnopharmacological studies revealed that the leaves and stems of Mimosa pudica L. (Fabaceae) are widely used for the treatment of epilepsy. This study sought to investigate the effects of the aqueous extract of Mimosa pudica leaves and stems against pilocarpine-picrotoxin kindling-induced temporal lobe epilepsy in mice and its implication on oxidative/nitrosative stress, GABAergic/cholinergic signalling, and brain-derived neurotrophic factor (BDNF) expression. The animals were treated for seven consecutive days as follows: one normal group and one negative control group that received orally distilled water; four test groups that received orally four doses of Mimosa pudica (20, 40, 80, and 160 mg/kg), respectively; and one positive control group that received 300 mg/kg sodium valproate intraperitoneally. One hour after the first treatment (first day), status epilepticus was induced by intraperitoneal injection of a single dose of pilocarpine (360 mg/kg). Then, 23 hours after the injection of pilocarpine to the mice, once again, they received their different treatments. Sixty minutes later, they were injected with a sub-convulsive dose of picrotoxin (1 mg/kg), and the anticonvulsant property of the extract was determined. On day 7, open-field, rotarod, and catalepsy tests were performed. Finally, the mice were sacrificed, and the hippocampi were isolated to quantify some biochemical markers of oxidative/nitrosative stress, GABAergic/cholinergic signalling, and BDNF levels in the hippocampus. Mimosa pudica extracts (160 mg/kg) significantly increased the latency time to status epilepticus by 70.91%. It significantly decreased the number of clonic and tonic seizures to 9.33 ± 1.03 and 5.00 ± 0.89, and their duration to 11.50 ± 2.07 and 6.83 ± 0.75 s, respectively. Exploratory behaviour, motor coordination, and catalepsy were significantly ameliorated, respectively, in the open-field, rotarod, and catalepsy tests. Pilocarpine-picrotoxin-induced alteration of oxidant-antioxidant balance, GABA-transaminase stability, acetylcholinesterase/butyrylcholinesterase activity, and neurogenesis were attenuated by the extract (80-160 mg/kg). This study showed that the aqueous extract of Mimosa pudica leaves and stems ameliorated epileptogenesis of temporal lobe epilepsy and could be used for the treatment of temporal lobe epilepsy.


CLIC5 promotes myoblast differentiation and skeletal muscle regeneration via the BGN-mediated canonical Wnt/β-catenin signaling pathway.

Myoblast differentiation plays a vital role in skeletal muscle regeneration. However, the protein-coding genes controlling this process remain incompletely understood. Here, we showed that chloride intracellular channel 5 (CLIC5) exerts a critical role in mediating myogenesis and skeletal muscle regeneration. Deletion of CLIC5 in skeletal muscle leads to reduced muscle weight and decreases the number and differentiation potential of satellite cells. In vitro, CLIC5 consistently inhibits myoblast proliferation while promoting myotube formation. CLIC5 promotes myogenic differentiation by activating the canonical Wnt/β-catenin signaling pathway in a biglycan (BGN)-dependent manner. CLIC5 deletion impairs muscle regeneration. Paired box gene 7 (Pax7) expression and the activity of BGN-mediated canonical Wnt/β-catenin signaling are reduced in CLIC5-deficient mice. Conversely, increasing CLIC5 levels in skeletal muscles enhances muscle regeneration capacity. In conclusion, our findings underscore CLIC5 as a pivotal regulator of myogenesis and skeletal muscle regeneration, functioning through interaction with BGN to activate the canonical Wnt/β-catenin signaling pathway.


Molecular evolution of nucleoside deoxyribosyl transferase to enhance the activity toward 2'-fluoro-2'-deoxynucleoside.

Nucleoside deoxyribosyl transferase (NDT) is an enzyme that catalyzes the transfer of purine and pyrimidine bases between 2'-deoxyribonucleosides and is widely used for synthesizing nucleoside analogs in various biotechnological applications. While NDT exhibits high activity toward natural nucleosides, its activity toward unnatural nucleoside analogs is significantly lower. Previously, the NDT mutant named fNDT(L59Q) was identified displaying 4.4-fold higher activity toward 2'-fluoro-2'-deoxyuridine (2FDU). In this study, molecular evolution strategies using error-prone PCR were employed to further generate mutant enzymes with enhanced activity toward 2FDU. After two rounds of mutational screening, two mutant clones that exhibited high activity against 2FDU were identified as fNDT-i1 (V52A) and fNDT-i2 (L28I), respectively. A double mutant, fNDT-i4, was subsequently constructed by combining the V52A and L28I mutations. Whole-cell-based activity measurements showed that fNDT-i4 exhibited 4.0- and 20.6-fold higher activity at 40°C and 50°C, respectively, compared to the wild-type NDT. The detailed characterization of the purified enzymes conducted under various conditions, including temperature, pH, thermal stability, and enzyme kinetics experiments, showed that fNDT-i1 and fNDT-i4 exhibited 3.1- and 3.7-fold higher catalytic efficiency, respectively than wild-type NDT. The L59Q mutation was identified as a key factor in improving the thermal stability, whereas the V52A and L28I mutations were critical for improving substrate affinity and reaction efficiency. These findings provide the potential of fNDT-i1 and fNDT-i4 as highly efficient biocatalysts for developing industrially relevant nucleoside analog synthesis.

 Acute Type A aortic dissection (TAAD) is a life-threatening condition that carries significant mortality and morbidity; a proportion of the survivors might require further aortic procedures in the mid-/long-term follow-up. Quality of life (QoL) after TAAD is not well studied. Quality of life after Type A Aortic Dissection Surgery (QUADS) is the first multicentre study to assess QoL in survivors of surgically treated TAAD.

Plant diseases caused by fungal pathogens are responsible for severe damage to strategic crops worldwide. Late wilt disease (LWD) is a vascular disease that occurs late in maize development. Harpophora maydis, the causative agent of maize LWD, is responsible for significant economic losses in Egypt. Therefore, the aim of this study was to control LWD of maize using an alternative approach to reduce the use of chemical pesticides. A combination of Trichoderma viride, a fungal biocontrol agent, and Azospirillum brasilense, a bacterial endophytic plant growth promoter, was applied in vitro and in planta. T. viride showed high mycoparasitic potential against H. maydis via various antagonistic activities, including the production of lytic enzymes, secondary metabolites, volatile compounds, and siderophores. A. brasilense and T. viride filtrates were also shown to suppress H. maydis growth, in addition to their ability to produce gibberellic and indole acetic acids. A significant change in the metabolites secreted by T. viride was observed using GC/MS in the presence of H. maydis. A field experiment was conducted on susceptible and resistant hybrids of maize to evaluate the antagonistic activity of T. viride combined with A. brasilense on LWD incidence as well as plant growth promotion under field conditions. The data revealed a significant decrease in both disease incidence and severity in maize plants treated with T. viride and/or A. brasilense. Further, there was a noticeable increase in all plant growth and yield parameters. An anatomical examination of the control and inoculated maize roots was also reflective of plant responses under biotic stress. Taken together, the obtained results provide successful eco-friendly management strategies against LWD in maize.


The importance of genotyping within the climate-smart plant breeding value chain - integrative tools for genetic enhancement programs.

The challenges faced by today's agronomists, plant breeders, and their managers encompass adapting sustainably to climate variability while working with limited budgets. Besides, managers are dealing with a multitude of issues with different organizations working on similar initiatives and projects, leading to a lack of a sustainable impact on smallholder farmers. To transform the current food systems as a more sustainable and resilient model efficient solutions are needed to deliver and convey results. Challenges such as logistics, labour, infrastructure, and equity, must be addressed alongside adapting to increasingly unstable climate conditions which affect the life cycle of transboundary pathogens and pests. In this context, transforming food systems go far beyond just farmers and plant breeders and it requires substantial contributions from industry, global finances, transportation, energy, education, and country developmental sectors including legislators. As a result, a holistic approach is essential for achieving sustainable and resilient food systems to sustain a global population anticipated to reach 9.7 billion by 2050 and 11.2 billion by 2100. As of 2021, nearly 193 million individuals were affected by food insecurity, 40 million more than in 2020. Meanwhile, the digital world is rapidly advancing with the digital economy estimated at about 20% of the global gross domestic product, suggesting that digital technologies are increasingly accessible even in areas affected by food insecurity. Leveraging these technologies can facilitate the development of climate-smart cultivars that adapt effectively to climate variation, meet consumer preferences, and address human and livestock nutritional needs. Most economically important traits in crops are controlled by multiple loci often with recessive alleles. Considering particularly Africa, this continent has several agro-climatic zones, hence crops need to be adapted to these. Therefore, targeting specific loci using modern tools offers a precise and efficient approach. This review article aims to address how these new technologies can provide a better support to smallholder farmers.
