Comments: The GFP variant in the plasmid backbone, pCSGFP2, contains the F64L and the S65T mutations resulting in 35 times brighter green emission (as described in Ikawa M et al., Curr. Top. Dev. Biol. 44 (1999), pp. 1–20) This construct contains the 5′ EcoRI fragment of the genomic clone XLF 4b (as described in Zapp D et al., Gene 134 (1993), pp. 251–256) linked to the EcoRI–HindIII fragment of the promoter clone 5Δ594 (as described in Zapp D et al., Mol. Cell. Biol. 13 (1993), pp. 6416–6426). By this strategy the sequence from −6 kbp to −58 bp upstream of the translation initiation codon of the HNF1α gene is linked to the GFP reporter present in pCSGFP2. Please note that there are some small discrepancies between Addgene's quality control sequence and the assembled sequence from the depositor. These discrepancies should not affect the function of the plasmid.
