Facultative Anaerobes Cultivation

Microorganisms can be categorized in several ways including aerobes (require oxygen to survive), facultative anaerobes (able to use oxygen for aerobic respiration but may also use fermentation or anaerobic respiration when oxygen is not present, or use fermentation in the presence of oxygen), or obligate anaerobes (cannot survive in the presence of oxygen). Some examples of facultatively anaerobic bacteria are Staphylococcus spp., Escherichia coli, Salmonella, Listeria spp., Shewanella oneidensis, and Yersinia pestis. Certain eukaryotes are also facultative anaerobes, including fungi such as Saccharomyces cerevisiae and many aquatic invertebrates such as nereid polychaetes. Culturing facultative anaerobes, however, poses a challenge for researchers because the equipment to grow, isolate, and identify them can be costly.

Aerobic Cultivation

Atmospheric condition is generally satisfactory for the culture of aerobes or facultative anaerobes but for the growth of many aerobes, it is necessary to provide extensive aeration. Forced aeration of cultures is therefore frequently desirable and can be achieved either by vigorously shaking the flask or tube on a shaker or by bubbling sterilized air into the medium. When aerobic organisms are to be grown in large quantities, it is advantageous to increase the exposure of the medium to the atmosphere. This can be accomplished by dispensing the medium in shallow layers or by providing aeration by constantly shaking the inoculated liquid cultures.

Isolation &amp; Cultivation of Specific Bacteria

Microorganisms exist in a mixed state in nature. To obtain the desired strains, they must be isolated. Accidental contamination during the preservation of the bacteria should also be purified. There are many methods of microbial isolation and purification, but the basic principle is similar, that is, the sample to be isolated is diluted to a certain extent, and the microbial cells (or spores) will exist in a dispersed state as far as possible, and then they will grow into a single colony of pure species. The above work is inseparable from inoculation, which is the process of transferring one microorganism to another sterilized medium. For the cultivation of specific bacteria, one or more special media suitable for the growth of specific bacteria are often used. The culture of bacteria, fungi, and actinomycetes is common, and the culture of single species can also be carried out.

Strain Anaerobic Culture

Anaerobic bacteria are widely distributed in nature, humans, and animals. Non-spore anaerobes mainly exist in human and animal bodies, especially in the intestines, oral cavity, upper respiratory tract, and urinary tract. Together with aerobic bacteria and facultative anaerobes, they form the body's normal flora. Anaerobic bacteria usually have an absolute advantage in normal flora. Under normal circumstances, the protection of the flora is relatively balanced, such as long-term use of broad-spectrum antibiotics, hormones, immunosuppressants, etc. However, the occurrence of flora imbalance or decreased body resistance can lead to endogenous anaerobic bacteria infection. The toxins produced by some anaerobic bacteria are extremely dangerous to advanced organisms including humans (such as tetanus toxin and botulinum toxin). Strain anaerobic culture refers to the culture of microorganisms in an oxygen-free environment. The techniques include the anaerobic tank method, anaerobic bag method, biological consumption method, pyrogallic acid method, and blister medium method, etc.
