Cell proliferation and migration are the determinants of malignant tumor progression, and a better understanding of related genes will lead to the identification of new targets aimed at preventing the spread of cancer. Some studies have shown that KIAA1199 (CEMIP) is a transmembrane protein expressed in many types of noncancerous cells and cancer cells. However, the potential role of KIAA1199 in the progression of cholangiocarcinoma (CCA) remains unclear.


TSPAN12 Precedes Tumor Proliferation by Cell Cycle Control in Ovarian Cancer.

TSPAN12, a member of the tetraspanin family, has been highly connected with the pathogenesis of cancer. Its biological function, however, especially in ovarian cancer (OC), has not been well elucidated. In this study, The Cancer Genome Atlas (TCGA) dataset analysis revealed that upregulation of TSPAN12 gene expression was significantly correlated with patient survival, suggesting that TSPAN12 might be a potential prognostic marker for OC. Further exploration showed that TSPAN12 overexpression accelerated proliferation and colony formation of OVCAR3 and SKOV3 OC cells. Knockdown of TSPAN12 expression in A2780 and SKOV3 cells decreased both proliferation and colony formation. Western blot analysis showed that several cyclins and cyclin-dependent kinases (CDK) (e.g., Cyclin A2, Cyclin D1, Cyclin E2, CDK2, and CDK4) were significantly involved in the regulation of cell cycle downstream of TSPAN12. Moreover, TSPAN12 accelerated mitotic progression by controlling cell cycle. Thus, our data demonstrated that TSPAN12 could be a novel molecular target for the treatment of OC.

Sequencing, Analysis and Organization of the Complete Genome of a Novel Baculovirus Calliteara abietis Nucleopolyhedrovirus (CaabNPV).

Baculoviridae, a virus family characterized by a single large double stranded DNA, encompasses the majority of viral bioinsecticides, representing a highly promising and environmentally friendly pesticide approach to insect control. This study focuses on the characterization of a baculovirus isolated from larvae of Calliteara abietis (Erebidae, Lymantriidae) collected in Mongolian pinaceae forests. This new isolate was called Calliteara abietis nucleopolyhedrovirus (CaabNPV). CaabNPV exhibits an irregular polyhedron shape, and significant variation in the diameter of its occlusion bodies (OBs) was observed. Nucleotide distance calculations confirmed CaabNPV as a novel baculovirus. The CaabNPV genome spans 177,161 bp with a G+C content of 45.12% and harbors 150 potential open reading frames (ORFs), including 38 core genes. A comprehensive genomic analysis categorizes CaabNPV within Group II alphabaculovirus, revealing a close phylogenetic relationship with Alphabaculovirus orleucostigmae (OrleNPV). Additionally, repeat sequence analysis identified three highly repetitive sequences consisting of 112 bp repeat units, known as homologous regions (hrs). This research contributes valuable insights into CaabNPV's phylogenetic placement, genomic structure, and its potential applications in insect biocontrol.

Nine taxa of the genus Pericalus Macleay from China are revised, with four new species described: Pericalus gibbosussp. n. (type locality: Putao, Myanmar; one paratype from Mêdog, Xizang), Pericalus eleganssp. n. (type locality: Mêdog, Xizang), Pericalus acutidenssp. n. (type locality: Longchuan, Yunnan), and Pericalus obscuratussp. n. (type locality: Fanjingshan, Guizhou). Four taxa are newly recorded from China: Pericalus ornatus ornatus Schmidt-Göbel (Yunnan, Hainan), P. obtusipennis Fedorenko (Yunnan), P. amplus Andrewes (Yunnan), and P. dux Andrewes (Yunnan). Pericalus formosanus Dupuis is newly ranked as a subspecies of P. ornatus Schmidt-Göbel. An improved key to world species of the subgenus Pericalus is provided, along with distribution maps and images of habitus and male and female genitalia for all Chinese species.

Arbuscular Mycorrhizal Symbiosis Alleviates Salt Stress in Black Locust through Improved Photosynthesis, Water Status, and K+/Na+ Homeostasis.

Soil salinization and the associated land degradation are major and growing ecological problems. Excess salt in soil impedes plant photosynthetic processes and root uptake of water and nutrients such as K+. Arbuscular mycorrhizal (AM) fungi can mitigate salt stress in host plants. Although, numerous studies demonstrate that photosynthesis and water status are improved by mycorrhizae, the molecular mechanisms involved have received little research attention. In the present study, we analyzed the effects of AM symbiosis and salt stress on photosynthesis, water status, concentrations of Na+ and K+, and the expression of several genes associated with photosynthesis (RppsbA, RppsbD, RprbcL, and RprbcS) and genes coding for aquaporins or membrane transport proteins involved in K+ and/or Na+ uptake, translocation, or compartmentalization homeostasis (RpSOS1, RpHKT1, RpNHX1, and RpSKOR) in black locust. The results showed that salinity reduced the net photosynthetic rate, stomatal conductance, and relative water content in both non-mycorrhizal (NM) and AM plants; the reductions of these three parameters were less in AM plants compared with NM plants. Under saline conditions, AM fungi significantly improved the net photosynthetic rate, quantum efficiency of photosystem II photochemistry, and K+ content in plants, but evidently reduced the Na+ content. AM plants also displayed a significant increase in the relative water content and an evident decrease in the shoot/root ratio of Na+ in the presence of 200 mM NaCl compared with NM plants. Additionally, mycorrhizal colonization upregulated the expression of three chloroplast genes (RppsbA, RppsbD, and RprbcL) in leaves, and three genes (RpSOS1, RpHKT1, and RpSKOR) encoding membrane transport proteins involved in K+/Na+ homeostasis in roots. Expression of several aquaporin genes was regulated by AM symbiosis in both leaves and roots depending on soil salinity. This study suggests that the beneficial effects of AM symbiosis on the photosynthetic capacity, water status, and K+/Na+ homeostasis lead to the improved growth performance and salt tolerance of black locust exposed to salt stress.

Risk stratification by long non-coding RNAs profiling in COVID-19 patients.

Coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has become a global pandemic worldwide. Long non-coding RNAs (lncRNAs) are a subclass of endogenous, non-protein-coding RNA, which lacks an open reading frame and is more than 200 nucleotides in length. However, the functions for lncRNAs in COVID-19 have not been unravelled. The present study aimed at identifying the related lncRNAs based on RNA sequencing of peripheral blood mononuclear cells from patients with SARS-CoV-2 infection as well as health individuals. Overall, 17 severe, 12 non-severe patients and 10 healthy controls were enrolled in this study. Firstly, we reported some altered lncRNAs between severe, non-severe COVID-19 patients and healthy controls. Next, we developed a 7-lncRNA panel with a good differential ability between severe and non-severe COVID-19 patients using least absolute shrinkage and selection operator regression. Finally, we observed that COVID-19 is a heterogeneous disease among which severe COVID-19 patients have two subtypes with similar risk score and immune score based on lncRNA panel using iCluster algorithm. As the roles of lncRNAs in COVID-19 have not yet been fully identified and understood, our analysis should provide valuable resource and information for the future studies.


Why does oriental arborvitae grow better when mixed with black locust: Insight on nutrient cycling?

To identify why tree growth differs by afforestation type is a matter of prime concern in forestry. A study was conducted to determine why oriental arborvitae (Platycladus orientalis) grows better in the presence of black locust (Robinia pseudoacacia) than in monoculture. Different types of stands (i.e., monocultures and mixture of black locust and oriental arborvitae, and native grassland as a control) were selected in the Loess Plateau, China. The height and diameter at breast height of each tree species were measured, and soil, shoot, and root samples were sampled. The arbuscular mycorrhizal (AM) attributes, shoot and root nutrient status, height and diameter of black locust were not influenced by the presence of oriental arborvitae. For oriental arborvitae, however, growing in mixture increased height and diameter and reduced shoot Mn, Ca, and Mg contents, AM fungal spore density, and colonization rate. Major changes in soil properties also occurred, primarily in soil water, NO 3-N, and available K levels and in soil enzyme activity. The increase in soil water, N, and K availability in the presence of black locust stimulated oriental arborvitae growth, and black locust in the mixed stand seems to suppress the development of AM symbiosis in oriental arborvitae roots, especially the production of AM fungal spores and vesicles, through improving soil water and N levels, thus freeing up carbon to fuel plant growth. Overall, the presence of black locust favored oriental arborvitae growth directly by improving soil water and fertility and indirectly by repressing AM symbiosis in oriental arborvitae roots.


The genus Pterostichus in China II: the subgenus Circinatus Sciaky, a species revision and phylogeny (Carabidae, Pterostichini).

All of the known species of the Chinese endemic subgenus Pterostichus (Circinatus) are revised, keyed, and illustrated. Eleven new species and one new subspecies are described: Pterostichus adelphus sp. n. (Sichuan: Meigu, N28.66°, E103.06°); Pterostichus ailaoicus sp. n. (Yunnan: Xinping, N23.94°, E101.50°); Pterostichus camelus sp. n. (Sichuan: Mianning, N28.97°, E102.16°); Pterostichus dimorphus sp. n. (Yunnan: Dayao, N26.08°, E101.03°); Pterostichus maitreya sp. n. (Guizhou: Fanjingshan, N27.90°, E108.70°); Pterostichus miao sp. n. (Guangxi: Maoershan, N25.87°, E110.41°); Pterostichus tumulus sp. n. (Guizhou: Fanjingshan, N27.90°, E108.70°); Pterostichus wangjiani sp. n. (Yunnan: Dongchuan, N26.08°, E102.87°); Pterostichus yan sp. n. (Hubei: Shennongjia, N31.47°, E110.39°); Pterostichus yuxiaodongi sp. n. (Sichuan: Wolong, N30.99°, E103.15°); Pterostichus zhygealu sp. n. (Sichuan: Meigu, N28.67°, E103.05°); and Pterostichus cavazzutianus mianningensis subsp. n. (Sichuan: Mianning, N28.97°, E102.16°). Pterostichus cavazzutianus is proposed as a replacement name for Pterostichus cavazuttii Allegro and Sciaky 2010, preoccupied by Pterostichus (Sinosteropus) barbarae cavazuttii Sciaky and Facchini 2003. A lectotype is designated for Pterostichus baenningeri Jedlička 1931. Two species, Pterostichus schuelkei Sciaky & Wrase and Pterostichus wenxianensis Allegro & Sciaky, are moved from the subgenus Circinatus to Morphohaptoderus. An infra-subgeneric taxonomy is proposed for the subgenus Circinatus with four species groups. The male endophallus characters for most species of Circinatus were well studied, with three types of endophallus defined. A phylogenetic analysis based on adult morphological characters confirmed the infra-subgeneric classification and clarified some of the relationships among species. Two main lineages within Circinatus were identified from the phylogenetic analyses. Three of the four species groups were monophyletic, whereas the fourth group was paraphyletic.


Taxonomic synopsis of the subtribe Physoderina (Coleoptera, Carabidae, Lebiini), with species revisions of eight genera.

Ten genera of Physoderina from the Oriental Region are diagnosed and described, and twenty six species representing eight genera (Paraphaea Bates, Anchista Nietner, Metallanchista gen. n., Diamella nom. n., Allocota Motschulsky, Orionella Jedlička, Endynomena Chaudoir and Dasiosoma Britton (Oriental species only)) are revised. Keys to genera and species are provided, along with distribution maps, habitus images, photographs of the name-bearing types, and illustrations of male and female genitalia of available species. The female internal reproductive system is illustrated for fourteen species. Two genera, Anchista and Taicona, previously placed in Calleidina, are moved into Physoderina. One new genus is described: Metallanchista, gen. n. (type species Metallanchista laticollis, sp. n.). Two new generic synonyms are proposed: Taicona Bates, 1873, junior synonym of Allocota Motschulsky, 1859; Teradaia Habu, 1979a, junior synonym of Dasiosoma Britton, 1937. A new generic replacement name is proposed: Diamella, nom. n. for Diamella Jedlička, 1952 (junior homonym of Diamella Gude, 1913). The status of Paraphaea Bates, 1873 is resurrected from synonym of Anchista Nietner, 1856. Five new species are described: Paraphaea minor Shi & Liang, sp. n. (Hoa-Binh, Tonkin, Vietnam), Anchista pilosa Shi & Liang, sp. n. (Chikkangalur, Bangalore, India), Metallanchista laticollis Shi & Liang, sp. n. (PhaTo env., Chumphon prov., Thailand), Allocota bicolor Shi & Liang, sp. n. (Dengga to Mafengshan, Ruili, Yunnan, China), Dasiosoma quadraticolle Shi & Liang, sp. n. (Menglun Botanical Garden, Yunnan, China). Fourteen new combinations are proposed: Paraphaea binotata (Dejean, 1825), comb. n. from Anchista; Paraphaea formosana (Jedlička, 1946), comb. n. from Anchista; Paraphaea philippinensis (Jedlička, 1935b), comb. n. from Allocota; Metallanchista perlaeta (Kirschenhofer, 1994), comb. n. from Allocota; Physodera andrewesi (Jedlička, 1934), comb. n. from Allocota; Diamella cupreomicans (Oberthür, 1883), comb. n. from Physodera; Diamella arrowi (Jedlička, 1935a), comb. n. from Allocota; Allocota aurata (Bates, 1873), comb. n. from Taicona; Dasiosoma bellum (Habu, 1979a), comb. n. from Teradaia; Dasiosoma indicum (Kirschenhofer, 2011), comb. n. from Diamella; Dasiosoma maindroni (Tian & Deuve, 2001), comb. n. from Lachnoderma; Dasiosoma hirsutum (Bates, 1873), comb. n. from Lachnoderma; Orionella discoidalis (Bates, 1892), comb. n. from Anchista; Orionella kathmanduensis (Kirschenhofer, 1994), comb. n. from Lachnoderma. One new replacement name is proposed: Dasiosoma basilewskyi, nom. n. for Dasiosoma hirsutum Basilewsky, 1949 (secondary junior homonym of Dasiosoma hirsutum (Bates, 1892)). One species is downgraded to subspecies rank: Anchista fenestrata subpubescens Chaudoir, 1877, new rank.


A comparative study of external female genitalia (including the 8 th and 9 th abdominal segments) in the family Megalopodidae and other related families of Chrysomeloidea.

The external female genitalia of 29 species belonging to three genera of Megalopodidae and 80 species belonging to 61 genera of another four families in Chrysomeloidea were studied. The external female genitalia within the superfamily Chrysomeloidea can be divided into a cerambycid type and a chrysomelid type. The comparative study of external female genitalia shows Megalopodidae is more closely related to the family Cerambycidae than to the family Chrysomelidae s.l. Among five subfamilies of Cerambycidae we studied, the subfamily Lamiinae is most closely allied to Megalopodidae. An evolutionary path is proposed for the spiculum gastrale in Chrysomeloidea: the characteristic state of the spiculum gastrale without a joint is primary, and that with a joint is secondary. The family Orsodacnidae has probably evolved in isolation from the early chrysomelids, due to their shared external female genitalia (cerambycid type). In the family Chrysomelidae, the structure of external female genitalia and ovipositing behavior show that the subfamily Synetinae is closer to the Camptosomata than the subfamily Eumolpinae. In general, the shape of the terminal ovipositor is palp-like in the Chrysomeloidea. Terminal ovipositors are generally palp-shaped in Chrysomeloidea except for those that are lamellate in the genus Callispa and the subfamily Cassidinae who produce egg-sheaths.


Identification of Transcriptional Variation in Aortic Remodeling Using a Murine Transverse Aortic Constriction (TAC) Model.

Arterial remodeling is a major pathological consequence of hypertension, which is recognized as the most common chronic non-communicable disease. However, the detailed mechanism of how arterial remodeling is induced by hypertension has not yet been fully elucidated. Evaluating the transcriptional changes in arterial tissue in response to elevated blood pressure at an early stage may provide new insights and identify novel therapeutic candidates in preventing arterial remodeling. Here, we used the ascending aorta of the transverse aortic constriction (TAC) model to induce arterial remodeling in C57BL/6 male mice. Age-matched mice were subjected to sham surgery as controls. The TAC model was only considered successful if the mice conformed to the criteria (RC/LC blood flow velocity with 5-10-fold change) 1 week after the surgery. Two weeks after surgery, the ascending aorta developed severe remodeling in TAC mice as compared to the sham group. High throughput sequencing was then applied to identify differentially expressed (DE) transcripts. In silicon analysis were then performed to systematically network transcriptional changes. A total of 1,019 mRNAs were significantly changed between TAC and the sham group at the transcriptional level. GO (Gene Ontology) and KEGG (Kyoto Encyclopedia of Genes and Genomes) analysis revealed that stress/stimulus/immune-related biological processes played a crucial role during arterial remodeling. Our data provide a comprehensive understanding of global gene expression changes in the TAC model, which suggests that targeting inflammation and vascular smooth cell transformation are potential therapeutic strategies to interfere with the aortic remodeling at an early stage in the development of hypertension.


NU6300 covalently reacts with cysteine-191 of gasdermin D to block its cleavage and palmitoylation.

Gasdermin D (GSDMD) serves as a vital mediator of inflammasome-driven pyroptosis. In our study, we have identified NU6300 as a specific GSDMD inhibitor that covalently interacts with cysteine-191 of GSDMD, effectively blocking its cleavage while not affecting earlier steps such as ASC oligomerization and caspase-1 processing in AIM2- and NLRC4-mediated inflammation. On the contrary, NU6300 robustly inhibits these earlier steps in NLRP3 inflammasome, confirming a unique feedback inhibition effect in the NLRP3-GSDMD pathway upon GSDMD targeting. Our study reveals a previously undefined mechanism of GSDMD inhibitors: NU6300 impairs the palmitoylation of both full-length and N-terminal GSDMD, impeding the membrane localization and oligomerization of N-terminal GSDMD. In vivo studies further demonstrate the efficacy of NU6300 in ameliorating dextran sodium sulfate-induced colitis and improving survival in lipopolysaccharide-induced sepsis. Overall, these findings highlight the potential of NU6300 as a promising lead compound for the treatment of inflammatory diseases.

Genome Analysis of a Novel Clade b Betabaculovirus Isolated from the Legume Pest Matsumuraeses phaseoli (Lepidoptera: Tortricidae).

Matsumuraeses phaseoli is a Lepidopteran pest that primarily feeds on numerous species of cultivated legumes, such as Glycine and Phaseolus. It is widely distributed in northeast Asia. A novel granulovirus, designated as Matsumuraeses phaseoli granulovirus (MaphGV), was isolated from pathogenic M. phaseoli larvae that dwell in rolled leaves of Astragalus membranaceus, a Chinese medicinal herb. In this study, using next-generation sequencing, we report the complete genome of MaphGV. MaphGV genome comprises a double-stranded DNA of 116,875 bp, with 37.18% GC content. It has 128 hypothetical open reading frames (ORFs). Among them, 38 are baculovirus core genes, 18 are lepidopteran baculovirus conserved genes, and 5 are unique to Baculoviridae. MaphGV has one baculovirus repeat ORF (bro) and three inhibitors of apoptosis proteins (iap), including a newfound iap-6. We found two atypical baculoviral homologous regions (hrs) and four direct repeats (drs) in the MaphGV genome. Based on phylogenetic analysis, MaphGV belongs to Clade b of Betabaculovirus and is closely related to Cydia pomonellagranulovirus (CpGV) and Cryptophlebia leucotretagranulovirus (CrleGV). This novel baculovirus discovery and sequencing are invaluable in understanding the evolution of baculovirus and MaphGV may be a potential biocontrol agent against the bean ravaging pest.
