A facile preparation method for new two-component supramolecular hydrogels and their performances in adsorption, catalysis, and stimuli-response.

In this study, we prepared a novel multifunctional two-component supramolecular hydrogel (T-G hydrogel) via two organic molecules in ethanol/water mixed solvents. In addition, we prepared gold nanoparticle/T-G (AuNPs/T-G) composite hydrogels using T-G hydrogel as a template for stabilizing AuNPs by adding HAuCl4 and NaBH4 during the heating and cooling process of T-G hydrogels. The morphology and microstructure of the as-prepared hydrogels were characterized using SEM, TEM, XRD, and FT-IR. The hydrogels prepared by solutions that contained different ethanol/water volume ratios exhibited different microstructures, such as sheets, strips, and rods. The obtained T-G hydrogels exhibited a sensitive response to pH changes in the process of sol-gel transformation and showed good adsorption properties for model organic dyes. In the presence of NaBH4, the obtained AuNP/T-G composite hydrogels exhibited the excellent catalytic performance for 4-nitrophenol (4-NP) degradation. Thus, the current research provides new clues in developing new multifunctional two-component supramolecular gel materials and exhibits potential applications for wastewater treatment.


Fabrication of Hydrogels via Host-Guest Polymers as Highly Efficient Organic Dye Adsorbents for Wastewater Treatment.

New self-assembled hydrogel materials of poly(vinyl alcohol)/cyclodextrin-modified poly(acrylic acid)/azobenzene-modified poly(acrylic acid) (PVA/PAA-CD/PAA-Azo) were successfully prepared via host-guest interactions and hydrogen bonds. The as-prepared hydrogel materials were characterized by various techniques, including Fourier transform infrared spectroscopy, X-ray diffraction analysis, scanning electron microscopy, ultraviolet spectroscopy, and specific surface area tests. The prepared hydrogels with different concentrations of PVA exhibited different network structures. In addition, ultraviolet (UV) light irradiation and temperature change induce a gel-sol phase transition in the hydrogel materials. The obtained hydrogel materials could be used as good adsorbents for two model organic dye molecules, which was mainly due to electrostatic interactions between methylene blue/rhodamine B (MB/RhB) and the gels in the adsorption process. In particular, the adsorption processes of the as-prepared hydrogel materials conformed to the pseudo-first-order model with a high correlation coefficient, which indicates that gel has a potential application in the field of wastewater purification.


Implantable PEKK/tantalum microparticles composite with improved surface performances for regulating cell behaviors, promoting bone formation and osseointegration.

Polyetherketoneketone (PEKK) exhibits admirable biocompatibility and mechanical performances but bioinert while tantalum (Ta) possesses excellent osteogenesis and osseointegration but high elastic modulus and density, and processing is too difficult and expensive. In the present study, combining of the advantages of both PEKK and Ta, implantable composites of PEKK/Ta were fabricated by blending PEKK with Ta microparticles of 20 v% (PT20) and 40 v% (PT40) content. In comparison with PT20 and PEKK, the surface hydrophilicity, surface energy, roughness and proteins adsorption as well as mechanical performances of PT40 significantly increased because of the higher Ta particles content in PEKK. Furthermore, PT40 exhibited the mechanical performances (e.g., compressive strength and modulus of elasticity) close to the cortical bone of human. Compared with PT20 and PEKK, PT40 with higher Ta content remarkably enhanced the responses (including adhesion, proliferation and osteogenic differentiation) of MC3T3-E1 cells in vitro. Moreover, PT40 markedly improved bone formation as well as osseointegration in vivo. In short, incorporation of Ta microparticles into PEKK created implantable composites with improved surface performances, which played key roles in stimulating cell responses/bone formation as well as promoting osseointegration. PT40 might have great potential for bear-loading bone substitute.

As an implantable biomaterial, polyetherketoneketone (PEKK) exhibits good mechanical strength but it is biologically inert while tantalum (Ta) possesses outstanding osteogenic bioactivity but has a high density and elastic modulus. Also, silicon nitride (SN) has osteogenic and antibacterial activity. In this study, a microporous surface containing both SN and Ta microparticles on PEKK (STP) exhibiting excellent osteogenic and antibacterial activity was created by sulfonation. Compared with sulfonated PEKK (SPK) without microparticles, the surface properties (roughness, surface energy, hydrophilicity and protein adsorption) of STP significantly increased due to the SN and Ta particles presence on the microporous surface. In addition, STP also exhibited outstanding antibacterial activity, which inhibited bacterial growth in vitro and prevented bacterial infection in vivo because of the presence of SN particles. Moreover, the microporous surface of STP containing both SN and Ta particles remarkably induced response (e.g., proliferation and differentiation) of rat bone mesenchymal stem (rBMS) cells in vitro. Furthermore, STP significantly improved new bone regeneration and osseointegration in vivo. Regarding the induction of cellular response in vitro and improvement of osseointegration in vivo, the microporous surface containing Ta was better than the surface with SN particles. In conclusion, STP with optimized surface properties activated cellular responses in vitro, enhanced osseointegration and prevented infection in vivo. Therefore, STP possessed the dual biofunctions of excellent osteogenic and antibacterial activity, showing great potential as a bone substitute.


Development of sandwich ELISA and lateral flow assay for the detection of Bungarus multicinctus venom.

Snakebite envenoming adversely affects human health and life worldwide. Presently, no suitable diagnostic tools for snakebite envenoming are available in China. Therefore, we sought to develop reliable diagnostic tests for snakebite management. We conducted affinity purification experiments to prepare species-specific antivenom antibody (SSAb). In brief, affinity chromatography with an antibody purification column (Protein A) was conducted to purify immunoglobulin G from Bungarus multicinctus (BM) venom hyperimmunized rabbit serum. The cross-reactive antibodies were removed from commercial BM antivenin by immune adsorption on the affinity chromatography columns of the other three venoms, Bungarus Fasciatus (FS), Naja atra (NA), and O. hannah (OH), generating SSAb. The results of western blot analysis and enzyme-linked immunosorbent assay (ELISA) showed the high specificity of the prepared SSAb. The obtained antibodies were then applied to ELISA and lateral flow assay (LFA) to detect BM venom. The resulting ELISA and LFA could specifically and rapidly detect BM venom in various samples with the limits of quantification as 0.1 and 1 ng/ml, respectively. This method could effectively detect snake venom in experimentally envenomed rats (simulating human envenomation), which could distinguish positive and negative samples within 10-15 min. This method also showed promise in serving as a highly useful tool for a rapid clinical distinguishing of BM bites and rational use of antivenom in emergency centers. The study also revealed cross-reactivity between BM and heterogenous venoms, suggesting that they shared common epitopes, which is of great significance for developing detection methods for venoms of the snakes belonging to the same family.
