Comments: Originally developed and characterized by Dr. Lester I. Binder at Northwestern University. The TOC1 antibody selectively recognizes oligomeric species of tau. The antibody was generated against cross-linked human tau dimers and originally the epitope was thought be a discontinuous epitope between amino acids 155-244 and 376-421 of full-length human tau (see Patterson et al., PMID: 21550980). However, more recent characterization of TOC1 indicates that the epitope is between aa 209-224 (see Ward et al., PMID: 23979027). Since the TOC1 epitope is continuous/linear, it must be used in non-denaturing assays such as dot blots, sandwich ELISAs and/or tissue immunostaining.
