The 17A2 monoclonal antibody specifically reacts with the mouse T lymphocytes receptor (TCR) associated CD3 complex, resulting in cellular activation and proliferation. CD3 is expressed by thymocytes and mature lymphocytes, and contains γ, δ, and ε subunits, involved in the assembly, trafficking, and surface expression of T-cell receptor complex. The interaction between the T lymphocytes and the 17A2 antibody can be blocked by the 145-2C11 anti-CD3e antibody, demonstrating that the 17A2 antibody recognizes the CD3 ε chain.

ProteoExtract Glycopeptide Enrichment Kit

Protein glycosylation influences crucial biological and pharmaceutically relevant events. Glycoanalysis studies of antibodies, receptors and other glycosylated proteins require highly sensitive, selective, efficient and reproducible methods for sample preparation and mass spectrometry (MS). Effective analysis requires glycopeptide enrichment to compensate for low abundance glycopeptides in complex mixtures and, subsequently, to enable glycan and attached peptide analysis by MS.The ProteoExtract Glycopeptide Enrichment Kit is designed to capture high-mannose, hybrid, and complex type glycosylated peptides from complex protein mixtures. The kit uses a proprietary ZIC Glycocapture Resin. In combination with the included optimized buffers, the ZIC Glycocapture Resin is highly selective for glycosylated peptides in the presence of abundant non-glycosylated peptides. The protocol is optimized to allow efficient removal of non-glycosylated peptides with an almost complete glycopeptide recovery (as quantitatively assessed by N-Glycan mapping). It allows both site-specific determination of the glycan structure and parallel analysis of the attached peptides. Glycopeptide enrichment is non-specific and nonselective, without preference for particular glycan structures and without loss of distinct glycans. The enriched glycopeptide fraction is directly compatible with downstream analysis by MALDI and ESI-MS. One ProteoExtract Glycopeptide Enrichment Kit provides sufficient reagents for 50 enrichment reactions. 10 µl sample with total protein concentration 2-4 mg/ml.

Simulated Extracellular Neural Recording Data

Background Extracellular recording of neural activity using microelectrode arrays is widely used in neuroscience research and invasive brain-computer interfaces. For detection of action potentials, or spikes, raw recordings are bandpass filtered and are often thresholded using an amplitude threshold. Computation of an optimal threshold is an active research area. ‘Truncation thresholdsare a pair of amplitude thresholds that are computed in an entirely automatic and data-driven manner for spike detection in filtered extracellular neural recordings (Okatan and Kocatürk, 2017). The software that computes them, theTruncation Thresholds Software’, is registered with SciCrunch.org under RRID:SCR_014637. Filtered recordings are widely accepted to consist of spikes and background noise. The maximum likelihood estimate of the standard deviation of the background noise is estimated by the Truncation Thresholds Software as a byproduct of the computation of the truncation thresholds. This estimate has been shown to be more accurate than the estimates obtained using seven other methods (Okatan, 2018a, 2018b). This result was demonstrated using simulated extracellular recording data. Here, the simulated data of Okatan (2018a, 2018b) are shared to stimulate further research in the development of new and more accurate estimators of the noise standard deviation. References M.
