The Iowa Gambling Task in fMRI images.

The Iowa Gambling Task (IGT) is a sensitive test for the detection of decision-making impairments in several neurological and psychiatric populations. Very few studies have employed the IGT in functional magnetic resonance imaging (fMRI) investigations, in part, because the task is cognitively complex. Here we report a method for exploring brain activity using fMRI during performance of the IGT. Decision-making during the IGT was associated with activity in several brain regions in a group of healthy individuals. The activated regions were consistent with the neural circuitry hypothesized to underlie somatic marker activation and decision-making. Specifically, a neural circuitry involving the dorsolateral prefrontal cortex (for working memory), the insula and posterior cingulate cortex (for representations of emotional states), the mesial orbitofrontal and ventromedial prefrontal cortex (for coupling the two previous processes), the ventral striatum and anterior cingulate/SMA (supplementary motor area) for implementing behavioral decisions was engaged. These results have implications for using the IGT to study abnormal mechanisms of decision making in a variety of clinical populations.


Survival of white-tailed deer fawns in central Iowa.

Understanding demographic parameters such as survival is important for scientifically sound wildlife management. Survival can vary by region, sex, age-class, habitat, and other factors. White-tailed deer fawn survival is highly variable across the species' range. While recent studies have investigated fawn survival in several Midwestern states, there have been no published estimates from Iowa for 30 years. We radio-collared 48 fawns in central Iowa from 2015-2017 to estimate survival, home range size, and habitat composition and identity causes of mortality. Estimated fawn survival (± SE) was similar to other Midwest studies at 30 (0.78 ± 0.07)) and 60 days (0.69 ± 0.08), but considerably lower at 7 months (0.31 ± 0.02). Survival was positively associated with woodland habitat through 30 and 60 days, but not related to habitat at 7 months. Female fawns avoided agricultural habitat in their home ranges. Fawn 95% kernel density home ranges were smaller than in other studies in the Midwest (21.22 ± 2.74 ha at 30 days, 25.47 ± 2.87 ha at 60 days, and 30.59 ± 2.37 ha at 7 months). The large amount of woodland and grassland (>90%) in our study area meant that fawns did not have to travel far to find suitable cover, which may explain their small home ranges. We recorded 21 mortalities, the leading cause of which was disease (n = 9; 56% epizootic hemorrhagic disease [EHD]) followed by suspected predation (4) and harvest (3). The mortality associated with an outbreak of EHD in 2016, all of which occurred after 60 days post-capture, is the most likely explanation for our low survival estimate at 7 months. While predation, usually early in life, is the leading cause of mortality in most studies, sporadic diseases like EHD can be a major source of mortality in older fawns in some years.


Farm residence and lymphohematopoietic cancers in the Iowa Women's Health Study.

Cancer incidence in male farmers has been studied extensively; however, less is known about risk among women residing on farms or in agricultural areas, who may be exposed to pesticides by their proximity to crop fields. We extended a previous follow-up of the Iowa Women's Health Study cohort to examine farm residence and the incidence of lymphohematopoietic cancers. Further, we investigated crop acreage within 750 m of residences, which has been associated with higher herbicide levels in Iowa homes.


Iowa Mutant Apolipoprotein A-I (ApoA-IIowa) Fibrils Target Lysosomes.

The single amino acid mutation G26R in human apolipoprotein A-I (apoA-IIowa) is the first mutation that was associated with familial AApoA1 amyloidosis. The N-terminal fragments (amino acid residues 1-83) of apoA-I containing this mutation deposit as amyloid fibrils in patients' tissues and organs, but the mechanisms of cellular degradation and cytotoxicity have not yet been clarified. In this study, we demonstrated degradation of apoA-IIowa fibrils via the autophagy-lysosomal pathway in human embryonic kidney 293 cells. ApoA-IIowa fibrils induced an increase in lysosomal pH and the cytosolic release of the toxic lysosomal protease cathepsin B. The mitochondrial dysfunction caused by apoA-IIowa fibrils depended on cathepsin B and was ameliorated by increasing the degradation of apoA-IIowa fibrils. Thus, although apoA-IIowa fibril transport to lysosomes and fibril degradation in lysosomes may have occurred, the presence of an excess number of apoA-IIowa fibrils, more than the lysosomes could degrade, may be detrimental to cells. Our results thus provide evidence that the target of apoA-IIowa fibrils is lysosomes, and we thereby gained a novel insight into the mechanism of AApoA1 amyloidosis.


Humor Improves Women's but Impairs Men's Iowa Gambling Task Performance.

The Iowa Gambling Task (IGT) is a popular method for examining real-life decision-making. Research has shown gender related differences in performance, in that men consistently outperform women. It has been suggested that these performance differences are related to decreased emotional control in women compared to men. Given the likely role of emotion in these gender differences, in the present study, we examine the effect of a humor induction on IGT performance and whether the effect of humor is moderated by gender. IGT performance and parameters from the Expectancy Valence Model (EVM) were measured in 68 university students (34 men; mean age 22.02, SD = 4.3 and 34 women; mean age 22.3, SD = 4.1) during a 100 trial-IGT task. Participants were exposed to a brief video before each of the IGT decisions available; one half of the samples (17 men and 17 women) was exposed to 100 humor videos, while the other half was exposed to 100 non-humor videos during the task. We observed a significant interaction between gender and humor, such that under humor, women's performance during the last block (trials 80-100) improved (compared to women under non-humor), whereas men's performance during the last block was worse (compared to men under non-humor). Consistent with previous work, under non-humor, men outperformed women in the last block. Lastly, our EVM results show that humor impacts the learning mechanisms of decision-making differently in men and women. Humor impaired men's ability to acquire knowledge about the payoff structure of the decks, and as a consequence, they were stuck in suboptimal performance. On the other hand, humor facilitated women's ability to explore and to learn from experience, improving performance. These findings deepen our understanding of the mechanisms underlying IGT decision-making and differential effects of humor in men and women.


Improving Iowa Research Network Patient Recruitment for an Advance Care Planning Study.

In February 2019, recruitment began in Iowa Research Network offices for a Patient-Centered Outcomes Research Institute (PCORI) funded Advance Care Planning (ACP) study to be conducted in 7 primary care practice-based research networks across the United States and Canada. The main study trained clinicians and nursing staff in serious illness care conversations and requested they refer eligible patients. Eligible patients were those with serious illness or frailty expected to live 1 to 2 years. Clinicians indicated it was difficult to identify eligible patients. This study aimed to find better methods for increasing patient recruitment for the ACP study.


Sustained effects of physical activity on bone health: Iowa Bone Development Study.

Studies of youth athletics and interventions have shown some maintenance of bone mineral content (BMC; g) after cessation of training, but less is known about sustained effects of everyday physical activity (PA). Using a prospective cohort, this report examined potential effects of childhood PA on adolescent BMC. Participants (N=156 boys, 170 girls) had exams at ages 5, 13, and 15. Body size and maturity were determined using anthropometry. Moderate-to-vigorous-intensity PA (MVPA) and vigorous-intensity PA (Vigorous PA) were measured using accelerometry. BMC of the spine and hip was measured using dual-energy X-ray absorptiometry. Mixed regression models tested whether PA at age 5 affected BMC at ages 13 and 15 after adjustment for age (year), height (cm), weight (kg), maturity (pre-peak height velocity or post), and activity level (min/day). Analysis was repeated to control for age 5 BMC. On average, boys participated in 59, 52, and 38 min of MVPA and 13, 17, and 11 min of Vigorous PA at ages 5, 13, and 15, respectively. MVPA (β=0.799) and Vigorous PA (β=1.338) at age 5 predicted later spine BMC (p<0.05). MVPA (β=0.480) at age 5 predicted hip BMC. Girls participated in 47, 33, and 26 min of MVPA and 10, 9 and 7 min of Vigorous PA at ages 5, 13, and 15, respectively. Neither MVPA nor Vigorous PA predicted later spine BMC. MVPA (β=0.302) at age 5 predicted hip BMC. After controlling for BMC at age 5 as well as the other covariates, the effect of MVPA (β=0.695) and Vigorous PA (β=1.079) at age 5 remained significant for boys at the spine. For girls, neither MVPA nor Vigorous PA at age 5 predicted spine or hip BMC. Children's early PA appears to have a modest effect on adolescent BMC at the critical regions of spine and hip; benefits may be greater for geometric changes, which future studies should include.


The orbitofrontal cortex represents advantageous choice in the Iowa gambling task.

A good-based model, the central neurobiological model of economic decision-making, proposes that the orbitofrontal cortex (OFC) represents binary choice outcome, that is, the chosen good. A good is defined by a group of determinants characterizing the conditions in which the commodity is offered, including commodity type, cost, risk, time delay, and ambiguity. Previous studies have found that the OFC represents the binary choice outcome in decision-making tasks involving commodity type, cost, risk, and delay. Real-life decisions are often complex and involve uncertainty, rewards, and penalties; however, whether the OFC represents binary choice outcomes in a complex decision-making situation, for example, Iowa gambling task (IGT), remains unclear. Here, we propose that the OFC represents binary choice outcome, that is, advantageous choice versus disadvantageous choice, in the IGT. We propose two hypotheses: first, the activity pattern in the human OFC represents an advantageous choice; and second, choice induces an OFC-related functional network. Using functional magnetic resonance imaging and advanced machine-learning tools, we found that the OFC represented an advantageous choice in the IGT. The OFC representation of advantageous choice was related to decision-making performance. Choice modulated the functional connectivity between the OFC and the superior medial gyrus. In conclusion, the OFC represents an advantageous choice during the IGT. In the framework of a good-based model, the results extend the role of the OFC to complex decision-making situation when making a binary choice.


Functional activity related to risk anticipation during performance of the Iowa Gambling Task.

Risk anticipation is an important cognitive/emotional component of decision making. The Iowa Gambling Task [Bechara, A., Damasio, A.R., Damasio, H., Anderson, S.W., 1994. Insensitivity to future consequences following damage to human prefrontal cortex. Cognition 50, 7-15], which is the most widely used "risk-anticipation task" in clinical studies, has been demonstrated to be sensitive to lesions involving the ventromedial prefrontal cortex or amygdala. However, the critical neural circuitry involved in this complex task has not yet been fully clarified even in healthy subjects. Using a 3-T scanner, we performed an event-related functional magnetic resonance imaging study in 14 healthy subjects performing the task. The statistical parametric mapping showed that the risk anticipation component (risky decisions minus safe decisions) exclusively activated the medial frontal gyrus. Furthermore, we found a significant interindividual correlation between the task performance and the magnitude of brain activity during risky decisions. These results indicate that the Iowa Gambling Task does recruit the neural circuitry that is critical in decision making under uncertainty, particularly when subjects perceive the risk of their decision.


New T2T assembly of Cryptosporidium parvum IOWA annotated with reference genome gene identifiers.

Cryptosporidium parvum is a significant pathogen causing gastrointestinal infections in humans and animals, that is spread through the ingestion of contaminated food and water. Despite its global impact on public health, generating a C. parvum genome sequence has always been challenging due to a lack of in vitro cultivation systems and challenging sub-telomeric gene families. A gapless telomere to telomere genome assembly has been created for Cryptosporidium parvum IOWA obtained from Bunch Grass Farms, named here as CpBGF. There are 8 chromosomes that total 9,259,183 bp. The new hybrid assembly which was generated with Illumina and Oxford Nanopore resolves complex sub-telomeric regions of chromosomes 1, 7 and 8. To facilitate ease of use and consistency with the literature, whenever possible, chromosomes have been oriented and genes in this annotation have been given the same gene IDs used in the current reference genome sequence generated in 2004. The annotation of this assembly utilized considerable RNA expression evidence, thus, untranslated regions, long noncoding RNAs and antisense RNAs are annotated. The CpBGF genome assembly serves as a valuable resource for understanding the biology, pathogenesis, and transmission of C. parvum, and it facilitates the development of diagnostics, drugs, and vaccines against cryptosporidiosis.


Low-level arsenic exposure from drinking water is associated with prostate cancer in Iowa.

Inorganic arsenic is a toxic naturally occurring element in soil and water in many regions of the US including the Midwest. Prostate cancer is the second most common type of cancer in men in Iowa, surpassed only by non-melanotic skin cancer. Epidemiology studies have evaluated arsenic exposure from drinking water and prostate cancer, but most have focused on high-level exposures outside the US. As drinking water from groundwater sources is a major source of arsenic exposure, we conducted an ecologic study to evaluate prostate cancer and arsenic in drinking water from public water sources and private wells in Iowa, where exposure levels are low, but duration of exposure can be long. Arsenic data from public water systems were obtained from the Iowa Safe Drinking Water Information System for the years 1994-2003 and for private wells from two Iowa Well Water Studies, the Iowa Community Private Well Study (ICPWS, 2002-2003) and Iowa Statewide Rural Well Water Survey Phase 2 (SWIRL2, 2006-2008) that provided data for 87 Iowa counties. Prostate cancer incidence data from 2009 to 2013 for Iowa were obtained from Surveillance, Epidemiology and End Results' SEER*Stat software. County averages of water arsenic levels varied from 1.08 to 18.6 ppb, with three counties above the current 10 ppb limit. Based on the tertiles of arsenic levels, counties were divided into three groups: low (1.08-2.06 ppb), medium (2.07-2.98 ppb), and high (2.99-18.6 ppb). Spatial Poisson regression modeling was conducted to estimate the risk ratios (RR) of prostate cancer by tertiles of arsenic level at a county level, adjusted for demographic and risk factors. The RR of prostate cancer were 1.23 (95% CI, 1.16-1.30) and 1.28 (95% CI, 1.21-1.35) in the medium and high groups, respectively, compared to the low group after adjusting for risk factors. The RR increased to 1.36 (95% CI, 1.28-1.45) in the high group when analyses were restricted to aggressive prostate cancers (Gleason score ≥ 7). This study shows a significant dose-dependent association between low-level arsenic exposure and prostate cancer, and if this result is replicated in future individual-level studies, may suggest that 10 ppb is not protective for human health.

Recollecting Cross-Cultural Evidences: Are Decision Makers Really Foresighted in Iowa Gambling Task?

The Iowa Gambling Task (IGT) has become a remarkable experimental paradigm of dynamic emotion decision making. In recent years, research has emphasized the "prominent deck B (PDB) phenomenon" among normal (control group) participants, in which they favor "bad" deck B with its high-frequency gain structure-a finding that is incongruent with the original IGT hypothesis concerning foresightedness. Some studies have attributed such performance inconsistencies to cultural differences. In the present review, 86 studies featuring data on individual deck selections were drawn from an initial sample of 958 IGT-related studies published from 1994 to 2017 for further investigation. The PDB phenomenon was found in 67.44% of the studies (58 of 86), and most participants were recorded as having adopted the "gain-stay loss-randomize" strategy to cope with uncertainty. Notably, participants in our sample of studies originated from 16 areas across North America, South America, Europe, Oceania, and Asia, and the findings suggest that the PDB phenomenon may be cross-cultural.


Aging and risky decision-making: New ERP evidence from the Iowa Gambling Task.

Several pieces of evidence have highlighted the presence of an age-related decline in risky decision-making (DM), but the reason of this decline is still unclear. The aim of the present study was to investigate the neural correlates of feedback processing in risky DM. Twenty-one younger (age <50 years) and 15 older (age >50 years) adults were tested with the Iowa Gambling Task (IGT) during Event Related Potentials (ERP) recording. The analysis was focused on the feedback-related negativity (FRN) and P3, two ERP components that represent different stages of feedback processing. Behavioral results revealed that older adults, despite showing a significant learning trend, completed the IGT with a gain of a smaller amount of money compared to the younger ones. ERP results revealed that while the FRN response was comparable in the two groups, the P3 amplitude was significantly reduced after negative feedback in older adults, compared with the younger ones. Furthermore, the difference in the P3 amplitude evoked by positive and negative feedback was significantly correlated with age. Hence, the present findings suggest that older adults seem to be less willing to shift attention from positive to negative information, and that this relevant change in the later stages of feedback processing could be the cause of a poor performance in risky DM contexts.


Sweating the small stuff: A meta-analysis of skin conductance on the Iowa gambling task.

To systematically examine the role of anticipatory skin conductance responses (aSCRs) in predicting Iowa Gambling Task (IGT) performance. Secondly, to assess the quality of aSCR evidence for the Somatic Marker Hypothesis (SMH) during the IGT. Finally, to evaluate the reliability of current psychophysiological measurements on the IGT. Electronic databases, journals and reference lists were examined for inclusion. Data were extracted by two reviewers and validated by another reviewer, using a standardised extraction sheet along with a quality assessment. Two meta-analyses of aSCR measures were conducted to test the relationship between overall aSCR and IGT performance, and differences in aSCR between advantageous and disadvantageous decks. Twenty studies were included in this review. Quality assessment revealed that five studies did not measure anticipatory responses, and few stated they followed standard IGT and/or psychophysiological procedures. The first meta-analysis of 15 studies revealed a significant, small-to-medium relationship between aSCR and IGT performance (r= .22). The second meta-analysis of eight studies revealed a significant, small difference in aSCR between the advantageous and disadvantageous decks (r= .10); however, publication bias is likely to be an issue. Meta-analyses revealed aSCR evidence supporting the SMH. However, inconsistencies in the IGT and psychophysiological methods, along with publication bias, cast doubt on these effects. It is recommended that future tests of the SMH use a range of psychophysiological measures, a standardised IGT protocol, and discriminate between advantageous and disadvantageous decks.


Tumor eosinophil infiltration and improved survival of colorectal cancer patients: Iowa Women's Health Study.

The role of the innate immune response in colorectal cancer is understudied. We examined the survival of colorectal cancer patients in relation to eosinophils, innate immune cells, infiltrating the tumor. Tissue microarrays were constructed from paraffin-embedded tumor tissues collected between 1986 and 2002 from 441 post-menopausal women diagnosed with colorectal cancer in the Iowa Women's Health Study. Tissue microarrays were stained with an eosinophil peroxidase antibody. Eosinophils in epithelial and stromal tissues within the tumor (called epithelial and stromal eosinophils, hereafter) were counted and scored into three and four categories, respectively. In addition, the degree of eosinophil degranulation (across epithelial and stromal tissues combined) was quantified and similarly categorized. We used Cox regression to estimate the hazard ratios and 95% confidence interval for all-cause and colorectal cancer death during 5-year follow-up after diagnosis and during follow-up through 2011 ('total follow-up'). The hazard ratios associated with eosinophil scores were adjusted for age of diagnosis, SEER (Surveillance, Epidemiology, and End Results) stage, tumor grade, body mass, and smoking history. High tumor stromal eosinophil score was inversely correlated with age and stage, and was associated with a decreased risk for all-cause and colorectal cancer death: hazard ratios (95% confidence intervals) were 0.61 (0.36-1.02; P-trend=0.02) and 0.48 (0.24-0.93; P-trend=0.01), respectively, during the 5-year follow-up for the highest vs lowest category. The inverse associations also existed for total follow-up for all-cause and colorectal cancer death for the highest vs lowest stromal eosinophil score: hazard ratios (95% confidence intervals) were 0.72 (0.48-1.08; P-trend=0.04) and 0.61 (0.34-1.12; P-trend=0.04), respectively. Further adjustment for treatment, comorbidities, additional lifestyle factors, tumor location, or molecular markers did not markedly change the associations, while adjustment for cytotoxic T cells slightly attenuated all associations. The infiltration of tumors with eosinophils, especially in stromal tissue, may be an important prognostic factor in colorectal cancer.


Salivary Antibodies against Multiple Environmental Pathogens Found in Individuals Recreating at an Iowa Beach.

Detecting environmental exposures and mitigating their impacts are growing global public health challenges. Antibody tests show great promise and have emerged as fundamental tools for large-scale exposure studies. Here, we apply, demonstrate and validate the utility of a salivary antibody multiplex immunoassay in measuring antibody prevalence and immunoconversions to six pathogens commonly found in the environment. The study aimed to assess waterborne infections in consenting beachgoers recreating at an Iowa riverine beach by measuring immunoglobulin G (IgG) antibodies against select pathogens in serially collected saliva samples. Results showed that nearly 80% of beachgoers had prior exposures to at least one of the targeted pathogens at the beginning of the study. Most of these exposures were to norovirus GI.1 (59.41%), norovirus GII.4 (58.79%) and Toxoplasma gondii (22.80%) and over half (56.28%) of beachgoers had evidence of previous exposure to multiple pathogens. Of individuals who returned samples for each collection period, 6.11% immunoconverted to one or more pathogens, largely to noroviruses (GI.1: 3.82% and GII.4: 2.29%) and T. gondii (1.53%). Outcomes of this effort illustrate that the multiplex immunoassay presented here serves as an effective tool for evaluating health risks by providing valuable information on the occurrence of known and emerging pathogens in population surveillance studies.

    1. Use quotes around phrases you want to match exactly
    2. You can manually AND and OR terms to change how we search between words
    3. You can add "-" to terms to make sure no results return with that term in them (ex. Cerebellum -CA1)
    4. You can add "+" to terms to require they be in the data
    5. Using autocomplete specifies which branch of our semantics you with to search and can help refine your search
