Comments: MCS is ClaI and SalI. This is a pro-retroviral vector for expression in chick tissue. It is based on the Pheonix system of Gary Nolan, Stanford University. The virus is packaged using Pheonix cells and VSVg pseudotyped. However, because of inclusion of a second, chicken promoter, it does not express well in mammalian cells. Can be titered using Dog osteosarcoma (D17) cells. Replication incompetent in chicken. Gives exceptionally bright GFP signal, suitable for live imaging and lineage tracing. This plasmid is unstable during transformation. If doing a fresh transformation, confirm correct size by digest (correct size is 6kb) before proceeding further.
