Description: 4Pi microscopy uses a special phase- and wavefront-corrected double-objective imaging system linked to a confocal scanner to enable 4-7 fold increased axial resolution over confocal & two photon microscopy. Even in living specimens, axial sections of ~100 nm are obtained. The system maintains all advantages of fast scanning, Acousto-Optical Beam Splitting (AOBS) and Spectral Detection of the Leica TCS SP2 AOBS for routine operation. The first marked leap in resolution in commercial 3D fluorescence microscopy opens up new dimensions for research in cell and developmental biology. The Leica TCS 4PI system was developed in close cooperation with Prof. Stefan Hell, Head of the NanoBiophotonics Department at the Max-Planck-Institute for Biophysical Chemistry, Goettingen, Germany and was in part funded by the German Ministry of Education and Research (BMBF).
