  • RRID:Addgene_11724

    This resource has 1+ mentions.

http://www.addgene.org/11724

Species: E. coli
Genetic Insert: b-lactamase
Vector Backbone Description: Backbone Marker:New England Biolabs; Backbone Size:2642; Vector Backbone:modified pGPS3; Vector Types:Bacterial Expression; Bacterial Resistance:Kanamycin
Defining Citation: PMID:11602576
Comments: Positive control for mutagenesis experiments: reversion of the stop codon results in ampicillin resistance. The pGPS3 has a deletion from BglI to BglII that destroys the ampicillin resistance gene. The ampicillin resistance gene containing the stop codon was then cloned in between AflIII and KpnI. Please also see pGFPuv from BD Biosciences as another control for forward mutation assays. Proper citation: RRID:Addgene_122486 Copy

http://www.addgene.org/113762

Species: E. coli
Genetic Insert: tdNfsB(F124W)-mCherry
Vector Backbone Description: Backbone Marker:Scott Sternson; Vector Backbone:AAV2; Vector Types:Mammalian Expression, AAV, Cre/Lox; Bacterial Resistance:Ampicillin
Defining Citation: PMID:30111097
Comments: Sternson Lab recommendations for propagation of AAV-FLEX plasmids: "These vectors are prone to recombination. This is a well known issue with these AAV vectors and is due to the inverted terminal repeats (ITRs) required for AAV production. To minimize recombination, we propagate these plasmids in NEB Stable cells. Also, to minimize recombination, cells should be cultured at 30 C. Note that these cultures will grow slowly (20 h for minipreps). Better yields and culture times are obtained with 2xYT as the media. This is strongly recommended. Because recombination may still happen occasionally, we do a panel of restriction digestions to assess whether the ITRs are in tact. Separate digestions with PvuII, Sma1, and SnaB1 should be performed."
